The Dysfunction of CD4+CD25+ Regulatory T Cells Contributes to the Abortion of Mice Caused by Toxoplasma gondii Excreted-Secreted Antigens in Early Pregnancy

Abstract: Toxoplasma gondii is an opportunistic intracellular parasite that is highly prevalent in human and warm-blooded animals throughout the world, leading to potentially severe congenital infections. Although the abortion caused by T. gondii is believed to be dependent on the timing of maternal infection during pregnancy, the mechanism remains unclear. This study was focused on the effects of T. gondii excreted-secreted antigens on pregnant outcomes and CD4+CD25+ Foxp3+ regulatory T cells at different stages of pre… Show more

“…To rule out the effect of ESA on the availability of IL-2, the concentration of IL-2 was assayed by ELISA after the administration of ESA. 8 Tregs are characterized by the constitutive expression of CD4, the IL-2 receptor α chain (CD25), and the specific transcription of Foxp3, which defines the development and suppressive function of Tregs. It indicated that ESA directly influences the level of IL-2Rγ.…”

“…9 T. gondii ESA could diminish the percentage of CD4 + CD25 + Foxp3 + in spleen and inhibit Foxp3 in the maternal-fetal interface. 8 Likewise, the administration of ESA reduced Foxp3 expression in EL4 cells. Our previous study revealed that ESA restricted transforming growth factor-β type II receptor (TGFβRII), phosphorylation of Smad2 (P-Smad2), phosphorylation of Smad3 (P-Smad3) and Smad4, subsequently leading to the downregulation of Foxp3.…”

“…Furthermore, during pregnancy, Tregs are key players in the tolerance toward the foetus bearing alloantigen . A link between the decreased number and the diminished suppressive capacity of Tregs has been linked to immunological rejection of the foetus, an effect that was partially abrogated by adoptive transfer of Tregs from normal pregnant mice into abortion‐prone animals . Foxp3 works as a master switch in modulating the development and function of Tregs, and it has been proposed that diminished expression of Foxp3 in endometrial tissue might be responsible for unexplained infertility .…”
