The Dynamic Control of Kiss-And-Run and Vesicular Reuse Probed with Single Nanoparticles

Abstract: Summary Vesicular secretion of neurotransmitter is essential for neuronal communication. Kiss-and-run is a mode of membrane fusion and retrieval without the full collapse of the vesicle into the plasma membrane and de novo regeneration. The significance of kiss-and-run during efficient neurotransmission has remained in doubt. We developed an approach for loading individual synaptic vesicles with single quantum dots. Their size and pH-dependent photoluminescence change allowed us to distinguish kiss-and-run fro… Show more

“…In dopaminergic neurons, the neurons vulnerable in Parkinson's disease, kiss-and-run-mediated exocytosis has been clearly shown [58,59]. However, in hippocampal neurons some studies argue that clathrin-mediated endocytosis is the main or even the only mechanism [54], while others report up to 60% of kiss-and-run endocytosis [60]. Clathrin-mediated endocytosis is one major mechanism of endocytosis, but there is still controversy on the involvement and relative importance of other mechanisms during 'normal' synaptic vesicle endocytosis, and it is not yet known if there are variations for different neuronal subtypes.…”

“…Different FM dyes exhibit distinct partitioning coefficients and wavelengths that allow for probing the system. Recently, quantum dots have been introduced as an alternative tracer to monitor fusion events 116 . A distinct advantage of such live tracers is the fact that they can be applied directly to preparations without a need for genetic manipulation or transfection.…”

“…This presents a challenge because synapses are small with complex nanoscale morphology which is not readily resolvable using conventional light microscopy. Elegant methods relying on nano-particles such as quantum dots 19,20 and super-resolution fluorescence imaging 8 provide important strategies to address this challenge, but are still limited in spatial information and ultrastructural context. An alternative approach is to combine a functional measure of vesicle pools with subsequent ultrastructural investigation.…”
