The dual role of curcumin and ferulic acid in counteracting chemoresistance and cisplatin-induced ototoxicity

Abstract: Platinum-based agents, such as cisplatin, form the mainstay of currently used chemotherapeutic regimens for several malignancies; however, the main limitations are chemoresistance and ototoxic side effects. In this study we used two different polyphenols, curcumin and ferulic acid as adjuvant chemotherapeutics evaluating (1) in vivo their antioxidant effects in protecting against cisplatin ototoxicity and (2) in vitro the transcription factors involved in tumor progression and cisplatin resistance. We reported… Show more

“…In fact, chemoresistance of cancer cells has urged scientists to seek new anti-tumor agents [ 261 ]. Based on the role of natural products in cancer treatment, they can be beneficial in sensitizing cancer cells into chemotherapy [ 262 , 263 ]. That is why these valuable agents have been extensively co-administered with chemotherapeutic agents in cancer therapy.…”

“…One thing to mention is that FA exhibits a biphasic response wherein at lower concentrations it exerts an oxidant function and at higher concentrations it promotes an antioxidant function for chemoresistance. Judging from this, curcumin seems the optimum regimen for effective treatment [ 223 ]. A novel synthetic polyphenol conjugate, (E)-3-(3,5-dimethoxyphenyl)-1-(2-methoxyphenyl) prop-2-en-1-one (DPP-23), can efficiently kill cisplatin-resistant HN3- cis R, HN4- cis R, and HN9- cis R cells without harming normal HOK-1 cells.…”

“…Increasing evidence demonstrates that phytochemicals, due to their excellent antitumor activity, can be considered as potential agents for coadministration with chemotherapeutic agents in cancer therapy [ 4 , 5 ]. Phytochemicals impair proliferation and metastasis of cancer cells, and sensitize cancer cells to apoptosis and DNA damage caused by chemotherapeutic agents [ 6 , 7 , 8 ]. This polychemotherapy with natural compounds is of importance in reversing chemoresistance.…”
