The Dual Hypocretin Receptor Antagonist Almorexant is Permissive for Activation of Wake-Promoting Systems

Abstract: The dual hypocretin receptor (HcrtR) antagonist almorexant (ALM) may promote sleep through selective disfacilitation of wakepromoting systems, whereas benzodiazepine receptor agonists (BzRAs) such as zolpidem (ZOL) induce sleep through general inhibition of neural activity. Previous studies have indicated that HcrtR antagonists cause less-functional impairment than BzRAs. To gain insight into the mechanisms underlying these differential profiles, we compared the effects of ALM and ZOL on functional activation … Show more

“…These data support the hypothesis that daridorexant selectively blocks orexin-induced excitation of wake-promoting regions but does not block the possible activation of these regions by other neurotransmitters released when one needs to wake up (Parks et al 2016 ). Positive GABA-A receptor modulators, in contrast, induce sleep through widespread inhibition of the CNS, and thus, the threshold for arousal might be increased.…”

“…S1f). The histamine‐ir pattern in AGS is identical to the pattern seen in rats stained with the same rabbit anti‐histamine () after 4% 1‐ethyl‐3‐[3‐dimethylaminopropyl] carbodiimide perfusion (Ellender et al, ; Noseda et al, ), and to the histaminergic neurons at bregma −3.80 identified by adenosine deaminase‐ir (Parks et al, ). TH‐ir was detected in the catecholaminergic neurons in the locus coeruleus (LC) (box1, Fig.…”
