The drosophila ortholog of the endolysosomal membrane protein, endolyn, regulates cell proliferation

Abstract: Endolyn (CD164) is a sialomucin that regulates the proliferation, adhesion, and migration of human haematopoietic stem and progenitor cells. This molecule is predominately localized in endocytotic compartments, where it may contribute to endolysosomal biogenesis and trafficking. In order to more closely define the function of endolyn from an evolutionary view-point, we first analyzed endolyn orthologs in species ranging from insects, fish, and birds to mammals. The predicted molecular structures of the endolyn… Show more

“…Previous work on Vsg revealed roles in promoting cell proliferation and embryonic development, though its specific function is unclear (Zhou et al, 2006). Its vertebrate homolog, endolyn, is targeted to endosomes and lysosomes, and functions in the maintenance of hematopoietic progenitors (Forde et al, 2007) and myoblast fusion (Bae et al, 2008).…”

“…In Drosophila , all existing loss of function alleles are homozygous viable. Although ablation of Vsg inhibits proliferation in S2 cells and loss-of-function mutations cause some reduction in embryonic viability, it seems otherwise not to be required for animal development and fecundity (Zhou et al 2006). The function of vsg in Drosophila hematopoiesis or immune response has not been previously studied.…”

“…While screening for suppressors of a LG overgrowth phenotype, three genes, visgun (vsg), SHC-adaptor protein (shc) and Adenosine deaminase-related growth factor A (Adgf-A), were identified where loss of function results in precocious plasmatocyte differentiation (Tan et al, 2012). vsg encodes an ortholog of mammalian endolyn, a endolysosomal sialomucin (Zhou et al, 2006) and shc encodes a SH2/PTB adaptor protein required for a subset of receptor tyrosine kinase receptors (Luschnig et al, 2000). Adgf-A expression is activated by JAK/ STAT signaling in the CZ, which lowers extracellular adenosine levels in the MZ, maintaining the prohemocyte population (Mondal et al, 2011(Mondal et al, , 2014.…”
