This study aims to analyze the condition of the coral reef ecosystem including the substrate, genus composition, and the level of coral mortality in the waters of the Malacca Strait, Masjid Raya District, Aceh Besar District. The research was conducted from October 2016 to October 2019 in the waters of the Malacca Strait, Masjid Raya District, Aceh Besar District. The research site is divided into three stations consisting of Lhok Mee Station, Ahmad Rhang Manyang and Benteng Inong Balee Station. Data were collected using the Point Intercept Transect method. The results showed that the condition of coral reefs at Lhok Mee Station was in the damaged category with coral cover ranging from 6.56% - 13.44%. Ahmad Rhang Manyang Station is included in the moderate to good category with coral cover ranging from 38.75% - 67.19%. Meanwhile, Benteng Inong Balee Station is included in the medium category with coral cover ranging from 45.94% - 56.88%. The monitoring results found that 30 coral genera with the highest composition of the coral genera came from the Porites genus, while other coral genera were unevenly distributed. The value of the mortality index for Lhok Mee Station shows a high increase in mortality of live coral to dead coral. Meanwhile, Ahmad Rhang Manyang and Benteng Inong Balee stations have low coral mortality index values.

Sponges have long been known as a source for isolating secondary metabolites. These natural compounds are biosynthetic products of symbiont bacteria from various phyla colonizing sponge tissue. Some symbiont bacteria are known to produce bioactive compounds that would have antibacterial activity, such as polyketide, due to competition in colonizing and obtaining nutrients from their hosts. In general, this study aims to explore the biosynthetic potential of seven sponge-symbiont bacteria by detecting the gene domain involved in the production of polyketide compounds. Sponge-symbiont bacteria isolation was carried out on one species of sponge collected from a depth of ±15 m in the Iboih area, Weh Island, Aceh Province, Indonesia. The bacteria was allowed to grow in Sea Water Complete agar medium and incubated at 280C for 10-14 days. The production of polyketide compounds involves the enzyme polyketide synthase (PKS). Polyketide synthase was detected by detecting the encoding gene domain involved in the production of polyketide compounds using PCR method. Five of the seven isolates of sponge symbiont bacteria were detected to contain the PKS gene domain. Furthermore, molecular identification confirm by 16S rRNA sequencing showed that the isolates belonged to the phylum Firmicutes and Actinobacteria. The result indicated that the sponge symbiont bacteria collected from Weh Island had the biosynthetic potential to produce polyketide compounds. These compounds would have antimicrobial activities that will play a major role in the medical field. Research related to screening PKS genes in marine sponge symbionts bacteria from Weh Island has never been reported before, thus adding to the novelty of this research.

The current research reports the length-weight relationships (LWRs) and Fulton’s condition factor (K) of three grouper species, namely: areolate grouper (Epinephelus areolatus), blacktip grouper (Epinephelus fasciatus), and honeycomb grouper (Epinephelus merra) harvested in the northern coast of Aceh, Indonesia. The fish samples were gathered from Pelabuhan Perikanan Samudera (PPS) Lampulo, Kuta Alam District, Banda Aceh, Aceh from June - December 2020. In total, 571 grouper specimens were collected during the study period (186 of E. areolatus, 205 specimens of E. fasciatus, and 180 E. merra). The total length (TL) of each fish species ranged from: 125.9-302.5 mm (mean 222.5 ± 30.3) (E. areolatus), 113.3-268.6 mm (mean 196.6 ± 26.9) (E. fasciatus) and 111.1-215.4 mm (mean 162.9 ± 21.7) (E. merra) with weight (W): 25.5-367.6 g (mean 145.8 ± 64.6) (E. areolatus), 24.0-290.9 g (mean 130.3 ± 56.2) (E. fasciatus) and 24.0-290.9 g (mean 130.3 ± 56.2) (E. fasciatus). In addition, the observed Fulton (K) condition factor and Ws values for all the three species were above 100 and 1. Furthermore, the LWRs analysis results revealed that all three fish species showed the b value of 3.0548 (r2=0.97), 3.0466 (r2=0.94) and 3.1681 (r2=0.97) for E. areolatus, E. fasciatus and E. merra, respectively, indicating that the three observed fish species had a positive allometric growth pattern and had a solid relationship between body weight and total length. This study delivers a reference point of several biological characteristics of the observed grouper species that will benefit and improve the fisheries management in the region.
