The diversity of conjugative relaxases and its application in plasmid classification

Abstract: Bacterial conjugation is an efficient and sophisticated mechanism of DNA transfer among bacteria. While mobilizable plasmids only encode a minimal MOB machinery that allows them to be transported by other plasmids, conjugative plasmids encode a complete set of transfer genes (MOB1T4SS). The only essential ingredient of the MOB machinery is the relaxase, the protein that initiates and terminates conjugative DNA processing. In this review we compared the sequences and properties of the relaxase proteins containe… Show more

“…pSmeSM11b of S. meliloti strain SM11 (Stiens et al, 2007). The predicted origin of conjugal transfer (oriT) of pSinA is placed between the traC and traA genes, and its location and nucleotide sequence (5'-CCAAGGGCGCAATTATACGTCGCTGACGCGACGTT-3') are conserved among plasmids belonging to the MOB Q2 family (Garcillán-Barcia et al, 2009). …”

“…A classification scheme has been proposed for fully sequenced plasmids based on phylogenetic analysis of the mobilization regions. Garcillan-Barcia et al, 2009;Smet et al, 2010;Smillie et al, 2010;Woodford et al, 2009) Characterization of plasmids on which ESBL and/or AmpC genes are located is an essential tool to study the epidemiology of these genes and plasmids. Since in Enterobacteriaceae often several different plasmids are present in each isolate, a structured approach is needed to identify the characteristics of the plasmid on which the β-lactamase genes are located.…”

“…Horizontal transmission of ICEs starts by their excision from the host chromosome through site-specific recombination and subsequent processing of the excised circularized DNA for conjugation, similar to conjugative plasmids (20,21) (Fig. 1A).…”
