The Discovery of Naringenin as Endolysosomal Two-Pore Channel Inhibitor and Its Emerging Role in SARS-CoV-2 Infection

Abstract: The flavonoid naringenin (Nar), present in citrus fruits and tomatoes, has been identified as a blocker of an emerging class of human intracellular channels, namely the two-pore channel (TPC) family, whose role has been established in several diseases. Indeed, Nar was shown to be effective against neoangiogenesis, a process essential for solid tumor progression, by specifically impairing TPC activity. The goal of the present review is to illustrate the rationale that links TPC channels to the mechanism of coro… Show more

“… 41 , 42 Moreover, naringin of citrus fruits has been shown to suppress the expression of proinflammatory cytokines during the inflammatory response. 41 , 43 Furthermore, hesperidin, a biologically active molecule in orange fruit, was shown to inactivate the SARS-CoV-2 RNA polymerase complex. 44 , 45 The antioxidant components of citrus fruits can also protect against oxidative stress associated with COVID-19 infection.…”

“…TPCs (Two Pore Channels) are cation channels involved in Ca 2+ signalling and activated by the ligands NAADP and PI(3,5)P 2 [24][25][26][27][28]. TPC proteins may be possible drug targets for the treatment of Ebola and SARS-CoV-2 viruses infection, since they control virus entry into the host cell [13,14,29,30]. Moreover, TPCs have been shown to be involved in neurodegenerative Parkinson's disease [31] and in processes of neoangiogenesis [32][33][34].…”

“…In TPC2-silenced Huh7.5 cells (silenced with siRNA), infection with the coronavirus HCoV 229E was inhibited significantly compared to the control, which indicates that TPC2 plays an active role in coronavirus infection. Since naringenin can inhibit TPC2, it has been proposed that TPC2 can be the molecular target of the compound [117].…”
