The development of fluorescence turn-on probe for Al(III) sensing and live cell nucleus-nucleoli staining

Abstract: The morphology of nucleus and nucleolus is powerful indicator of physiological and pathological conditions. The specific staining of nucleolus recently gained much attention due to the limited and expensive availability of the only existing stain “SYTO RNA-Select”. Here, a new multifunctional salen type ligand (L1) and its Al3+ complex (1) are designed and synthesized. L1 acts as a chemosensor for Al3+ whereas 1 demonstrates specific staining of nucleus as well as nucleoli. The binding of 1 with nucleic acid i… Show more

“…The nucleolus shows a grainy texture in Fig. 7(c), which conforms with previous works visualizing MCF-7 nucleoli via fluorescence [30]. The surrounding cytoplasm also exhibits significantly increased contrast, and various cytoplasmic extensions and cavities are clearly visible in sharp focus in Fig.…”

“…The binding mode of TPySSB with Al 3+ was also studied by analyzing electrospray ionization MS spectra. A characteristic peak at m/z 580.0779, corresponding to [TPySSB + 2Al 3+ + NO 3 − + 2H 2 O -H + ] + , 23 is present in the positive-ion mass spectrum of TPySSB-Al 3+ complex, confirming that the stoichiometry ratio between TPySSB and Al 3+ is 1:2. Figure 9 shows the 1 H NMR spectra of single TPySSB and complex TPySSB-Al 3+ .…”

“…The nucleolus is the site for rRNA transcription and for ribosomal subunit assembly . To determine the interaction between HL1 and RNA, we performed RNase digestion tests in HeLa cells, after incubating with HL1 and zinc salts , , . the RNase enzyme hydrolyzes RNA, which is present in the cells.…”
