This study was conducted to investigate the prophylactic effects of lycopene (LC) and ellagic acid (EA) on 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-induced testicular and spermatozoal toxicity. These toxicological changes are associated with the oxidative stress and apoptosis in male rats. Forty-eight male rats were allocated to one of six groups of 8 rats each: control, LC, EA, TCDD, TCDD+LC, and TCDD+EA. The control group was treated with 0.5 mL/rat slightly alkaline solution+0.5 mL/rat corn oil every other day. The LC group was treated with 0.5 mL/rat slightly alkaline solution+0.5 mL/rat corn oil containing 10 mg/kg of LC every other day. The EA group received 0.5 mL/rat corn oil+0.5 mL/rat slightly alkaline solution containing 2 mg/kg of EA every other day. The TCDD group received 0.5 mL/rat corn oil containing 100 ng/kg/day of TCDD+0.5 mL/rat slightly alkaline solution. The TCDD+LC group was treated with 0.5 mL/rat TCDD+0.5 mL/rat LC. The TCDD+EA group was treated with 0.5 mL/rat TCDD+0.5 mL/rat EA. All treatments were made by gavage, and the experimental period was maintained during 8 weeks. Sperm motility, concentration, and abnormal sperm rate in epididymal tissue, testicular tissue lipid peroxidation (LPO), antioxidant enzyme activity, histopathological changes, and apoptosis (i.e., Bax and Bcl-2 proteins) were determined. TCDD exposure resulted in significant decreases in sperm motility, concentration, testicular superoxide dismutase activity, germinal cell-layer thickness, Johnsen's testicular score, and significant increases in abnormal sperm rate, testicular malondialdehyde, glutathione levels, Bax-positive staining, and Bax-positive apoptotic cell score, along with some testicular histopathological lesions. TCDD treatment did not affect significantly catalase activity. However, combined treatment with LC or EA, in addition to TCDD, prevented the development of TCDD-induced damages in sperm quality, testicular histology, and LPO. Improvements in testicular apoptosis after the administration of LC and EA to TCDD-treated rats were minimal, but not statistically significant. TCDD-induced lipid peroxidation leads to functional and structural damages, as well as apoptosis, in spermatogenic cells of rats. Both LC and EA protected against the development of these effects.

Monitoring gonad maturation for protandrous and functional hermaphrodite species such as the giant clam Tridacna maxima is difficult due to the juxtaposition and relative proportion of male and female tissues in the gonad [gonadal sex ratio (GSR)].Here, the relevance of the widely used gonadosomatic index (GSI) as proxy of giant clam gonad maturation is tested with a large dataset (n ¼ 265). Gonadosomatic index is compared with other indices, namely the proportion of the male part harboring spermatozoids, the proportion of empty oocyte follicles, the mean oocyte diameter, and the oocyte elongation. At gonad scale, high index variability highlighted partial spawning. At individual scale, male and female maturation proxies were contrasted, showing either asynchronous emissions of male and female gametes or contrasted spermatogenesis and oogenesis duration. The GSI was mostly driven by the number and diameter of oocytes and therefore it is recommended here as primary proxy for female maturity. Except for the oocyte elongation, all indices were affected by the GSR, which ruled out drawing conclusions at population scale. These results highlight the need for maturation stage proxies that are optimized for functional hermaphrodite species.

Successful reintroduction of "ecologically extinct" bivalve species into anthropogenically impaired urban estuaries is problematic when employing existing management tools used in estuaries where bivalves are present (GIS-based restoration models, expanding existing shellfish beds, placement of shell substrate, physical oceanographic parameters). A significant management challenge is appropriate site selection. We are proposing the inclusion of a biological parameter (evaluation of tissue histopathology) in an inexpensive and rapid site selection model to inform management decision making and identify sites with the greatest potential for reintroduction success. Use of biological biomarkers is not a new concept, but it is important that they be included in a multitiered management approach to bivalve reintroduction. This Case Study tested adult Eastern Oysters (Crassostrea virginica Gmelin) from locations that supported comparable short-term survival rates by evaluating growth and tissue health and/or disease. Biomarkers indicated oyster tissues at one site were normal, the female:male sex ratio was 50:50, and female oysters were in spawning condition. Conversely, oyster tissues at the second site exhibited multiple abnormalities, samples were 100% male, and the incidence of disease was high. Using the biomarker tool, we evaluated 4 additional sites where oysters exhibited short-term (1 year) survival. At 2 locations, we observed chronic health impacts that would preclude reintroduction, including samples from one site where a wild population was surviving. We also analyzed tissue and shell heavy metal contents. Soft tissue metal concentrations in Meadowlands samples were at the high range of scientific literature values, averaging 1.1% of total body weight, whereas tissue metal concentrations at the Keyport site were within acceptable ranges. Although initial survival and growth rates at both locations were comparable, site-specific urban stressors reduced oyster fitness at 1 of the 2 locations. We are proposing an Estuarine Reintroduction Site Selection Model, which includes a biological in situ parameter, to increase the probability of successfully managing a sustainable oyster reintroduction before commencing expensive large-scale restoration activities.
