High antigenic variability in the envelope (E) protein of different virus strains has been a major obstacle in designing effective vaccines for Dengue virus (DENV). To maintain their biological function, some parts of viral proteins remain stable during evolution thus one possible approach to solve this problem is to recognize specific regions within different protein sequences of E that have the tendency to stay constant through evolution. These regions may possess some special attributes to become a vaccine candidate against dengue virus. In this study, a computational approach was utilized to identify and analyze highly conserved amino acid sequences of the DENV E protein. Sequences of 9 amino acids or more were specifically focused due to their immune-relevant as T-cell determinants. Different bioinformatics tools were responsible for revealing conserved regions in the DENV E protein and constructing the phylogenetic tree from the sequence database. The tools also predicted immunogenicity of the identified vaccine targets. Ultimately, two peptide regions of at least 9 amino acids were chosen due to their high conserved attribute in more than 95% of all collected DENV sequences. Moreover, both of them was found to be immune-relevant by their correspondence to known or putative HLA-restricted T cell determinants. The conserved attribute of these sequences through the entire analysis of this study supports their potential as candidates for further in vitro experiments for rational design a universal vaccine which has longer and broader impact.

The development of vaccines against flaviviruses, including Zika virus (ZIKV) and dengue virus (DENV), continues to be a major challenge, hindered by the lack of efficient and reliable methods for screening neutralizing activity of sera or antibodies. To address this need, we previously developed a plasmid-based, replication-incompetent DENV reporter virus particle (RVP) production system as an efficient and safe alternative to the Plaque Reduction Neutralization Test (PRNT). As part of the response to the 2015-2016 ZIKV outbreak, we recently developed pseudoinfectious ZIKV RVPs by modifying our DENV RVP system. The use of ZIKV RVPs as critical reagents in human clinical trials requires their further validation using stability and reproducibility metrics for large-scale applications. In the current study, we validated ZIKV RVPs using infectivity, neutralization, and enhancement assays with monoclonal antibodies (MAbs) and human ZIKV-positive patient serum. ZIKV RVPs are antigenically identical to live virus based on binding ELISA and neutralization results and are nonreplicating based on the results of plaque formation assays. We demonstrate reproducible neutralization titer data (NT50 values) across different RVP production lots, volumes, time frames, and laboratories. We also show RVP stability across experimentally relevant time intervals and temperatures. Our results demonstrate that ZIKV RVPs provide a safe, rapid, and reproducible reagent for large-scale, long-term studies of neutralizing antibodies and sera, which can facilitate large-scale screening and epidemiological studies to help expedite ZIKV vaccine development.

Larvicidal Properties of Citrus hystrix DC Fruit (Taw-Shaut-Khar-Thee) Extracts against Aedes aegypti Larvae Collected from Different Townships of Yangon

The present study aimed to evaluate the larvicidal and ovicidal activities of ethanol extracts of dried fruits, internal fruit materials and peels of Citrus hystrix DC against Aedes aegtpti larvae from Dagon (North) and Hlinethaya Townships in Yangon Region from May 20l5 to December 2016. The 3rd and 4th stage larvae were exposed for 24 hours to various concentrations of ethanol extracts of different parts of the Citrus hystrix fruit. The dry fruit extract resulted in significantly higher mortality of 100% and 96.4% in Aedes larvae collected from Dagon (North) and Hlinethaya, respectively (P<0.05). The mortality of Aedes larvae in peel extract was found to be 92.5% in Hlinethaya and 94.4% in Dagon (North), respectively. The extract of Citrus hystrix DC fruit was very effective to kill Aedes larvae collected from Dagon (North). When compared with each other, slightly higher mortality of larvae from Dagon (North) was observed at 0.l5 g/100 m1 concentration of all extracts than that of larvae from Hlinethaya. Ovicidal test found 100% protection to lay their eggs at 0.0l25 g/l00 ml dose and persistency test found 100% mortality of 3 rd and 4 th instar Aedes larvae up to 6 days at a dose of 0.2 g/100 ml fruit extract. The LC 50 and LC 90 values of 0.0343 g, 0.0415 g, 0.0231 g, and 0.1256 g, 0.1584 g, 0.0742 g for peel, internal material and fruit extracts, respectively, on Aedes larvae from Dagon were found to be lower than 0.0396 g, 0.0552 g, 0.0280 g and 0.1408 g, 0.2322 g, 0.1060 g, respectively, from Hlinethaya. The findings of the present study revealed that the ethanol extract of the fruit of Citrus hystrix DC has strong larvicidal, ovicidal and long-term persistency activity on Aedes mosquitoes as a good source of preparations for mosquito control. The study may contribute to a reduction in the application of synthetic insecticides, which in turnincreases the opportunity for natural control of various medically important insects by botanical insecticides.
