The Dectin 1 Agonist Curdlan Regulates Osteoclastogenesis by Inhibiting Nuclear Factor of Activated T cells Cytoplasmic 1 (NFATc1) through Syk Kinase

Abstract: Background: Dectin 1 is found on myeloid lineage cells and contains an immunoreceptor tyrosine-based activation motif from which signals are associated with bone homeostasis. Results: The dectin 1 agonist curdlan suppresses osteoclastogenesis induced by receptor activator of NF-B ligand (RANKL). Conclusion: Curdlan regulates RANKL-induced osteoclastogenesis.Significance: Curdlan could be a potential therapeutic candidate in treating osteoclast-related diseases.

“…β-1-3-glucans are agonist ligands of dectin-1 and present on the cell wall of various fungal pathogens, such as Candida albicans [ 15 17 ]. Previous studies indicate that the dectin-1 agonist curdlan suppresses RANKL-induced osteoclastogenesis [ 18 ]. However, the mechanisms underlying its anti-osteoclastogenic effects need to be further elucidated.…”

“…1B). As resveratrol and piceatannol are reported to have anti-osteoclastogenic activities (31,32) and dRG exhibited potent anti-osteoclastogenic activity, the present study further investigated the anti-osteoclastogenic effect of dRG.…”

“…TAS05567 Suppresses Development of TRAP-Positive Multinucleated Cells. In addition to its crucial role in inflammation, Syk contributes directly to bone resorption in RA via its kinase activity-mediated regulation of receptor activator of nuclear factor kB (RANK) signaling pathway that is essential for differentiation and activation of osteoclasts (Yamasaki et al, 2014). Therefore, we explored the effect of TAS05567 on osteoclast differentiation using human osteoclast precursor cells.…”
