The cytotoxin α‐sarcin behaves as a cyclizing ribonuclease

Abstract: The hydrolysis of adenylyl(3PC5P)adenosine (ApA) and guanylyl(3PC5P)adenosine (GpA) dinucleotides by the cytotoxic protein K K-sarcin has been studied. Quantitative analysis of the reaction has been performed through reversephase chromatographic (HPLC) separation of the resulting products. The hydrolysis of the 3P-5P phosphodiester bond of these substrates yields the 2P-3P cyclic mononucleotide; this intermediate is converted into the corresponding 3P-monophosphate derivative as the final product of the reacti… Show more

“…No effect on vesicles composed of zwitterionic phospholipids has been detected. It was assumed that the antifungal activity of AFP is related to its ability to interact with anionic phospholipids (13).…”

“…However, they can also produce extensive non-specific digestion of almost any RNA when used at higher concentrations [9,41]. Ribotoxins follow the same general acid--base type endonucleolytic reaction scheme as the other members of the RNase T1 family [2,3,42,43]. of the RNase T1 family so far studied is implicit in the denomination of all these enzymes as cyclizing RNases [43].…”
