The Curcumin Analog EF24 Targets NF-κB and miRNA-21, and Has Potent Anticancer Activity In Vitro and In Vivo

Abstract: EF24 is a curcumin analog that has improved anticancer activity over curcumin, but its therapeutic potential and mechanism of action is unknown, which is important to address as curcumin targets multiple signaling pathways. EF24 inhibits the NF-κB but not the JAK-STAT signaling pathway in DU145 human prostate cancer cells and B16 murine melanoma cells. EF24 induces apoptosis in these cells apparently by inhibiting miR-21 expression, and also enhances the expression of several miR-21 target genes, PTEN and PDCD… Show more

“…42 miRNAs can be modulated by botanicals in lung cancer. [43][44][45][46] The effects of curcumin on a human retinal pigment epithelial cell line, ARPE-19, included downregulation of 20 and upregulation of 9 miRNAs. 47 Effects of resveratrol on miRNA have also been studied.…”

“…Curcumin could function as a selective regulator of the NF-κB signaling to block the activation of the NF-κB which resulted in the inhibition of miR-21 expression. 29,30 Methylation as a fundamental regulator of gene transcription may be another mechanism by which curcumin regulates miRNA expression. For example, curcumin could cause epigenetic modulation of miR-203 expression through demethylating the miR-203 promoter in bladder cancer.…”

“…Therefore, by downregulating survivin, PG might also exert an antimetastatic effect in OSCC in addition to inducing apoptosis. microRNAs (miRNAs) are short noncoding RNAs that regulate the expression of multiple genes at the posttranscriptional level [35] . Numerous studies have established that dysregulation of miRNA expression is present in various human tumors [36] .…”
