Híbrido de Timor is the principal source for disease and pest resistance genes in C. arabica breeding program worldwide. The part of the chromosome responsible for resistance introgressed from C. canephora to Híbrido de Timor are claimed to affect the cup quality of the C. arabica cultivars derived from the crossing program of Híbrido de Timor. Therefore, this work was done to study the genome introgression of Híbrido de Timor and its impact on the cup quality on the C. arabica cultivars. For genome introgression and genetic relationship analysis, seventy-six accessions from C. arabica, C. canephora and Híbrido de Timor were analyzed using AFLP and SSR molecular markers. To understand the effect of genome introgressed from Híbrido de Timor on cup quality, three C. arabica, seven Híbrido de Timor and six cultivars derived from Híbrido de Timor × C. arabica were genotyped using SSR molecular markers and sensorial analysis was performed. The genetic diversity analysis among the tested genotypes showed high genetic similarity between Híbrido de Timor with C. arabica and clear differentiation among coffee species. The analysis of genome introgression of C. arabica and C. canephora var Robusta into Híbrido de Timor not reach 30% of C. canephora genome. The sensorial analysis of coffee genotypes demonstrated non-significant difference on cup quality parameters among C. arabica cv Bourbon and cultivars derived from Híbrido de Timor that showed the possibility of developing C. arabica cultivars without affecting the cup quality. Similarly, the SSR marker diversity showed high genetic similarity between the Bourbon and the C. arabica cultivars derived from Híbrido de Timor.

Development of specialty coffee product needs to be done by producing a product that has a good flavor, unique, and different from the axisting specialty coffee products. One of the varieties of Arabica coffee that has good potential to be developed into a specialty coffee is Maragogype coffee. This paper discusses the results of selection and characterization of physical and flavor quality of Arabica coffee varieties Maragogype in East Java. Selection was done in PTPN XII at Pancur/Angkrek and Kayumas Estate. It was obtained two superior genotypes those have high productivity and good taste, they are MP3 and MP4 with of productivity of 7,985.3 and 5,985.3 g cherries/tree, respectively. Maragogipe varieties showed good physical quality, in which 99% of bean belong to the large been category, whiches the highest of 1,000 beens, and good bulk density and apparent swelling. MP3 genotype had a floral, citrus acidity, mild, and very good sweetness characteristics. MP mix had the showed same characteristics, but its intensity was not as high as MP3. MP4 has herbal and flora characteristics with bitter after taste.Key words : Coffea arabica, maragogype, specialty coffee, characterization, selection, flavor, variety.

Canopy position influences on gene expression and coffee quality

Coffee is one of the most valuable commodities exported around the world. Only two out of 125 species are highly valued in the genus Coffea, Arabica and Robusta coffee, accounting for 62% and 35% of the world coffee production. This project aimed to improve the understanding of the molecular and genetic basis of coffee quality through the detection of the upper and lower canopy position influences on coffee bean quality and gene expression. Selection from different parts of the plant might help to meet the increased threat from climate change and the demands of highly discerning consumers. Coffee quality is a very complicated trait depending on different perspectives. Phenotypic analysis was used to systematically access coffee quality through measurement of physical traits (including bean weight and size (length, width and thickness)), chemical components analysis (including determination of sucrose, trigonelline and caffeine content) and sensory evaluation to assess the aroma. To understand the influence of canopy position on coffee quality and gene expression, transcriptome analysis was conducted in this study. A reference transcriptome was generated for Arabica coffee using long read sequencing. To understand the dynamic of canopy position influences, three different ripening stages were selected. Upper canopy samples were used to understand the processes associated with coffee bean ripening and how the key components were accumulated in coffee beans through ripening. Thereafter, comparative transcriptome analysis was conducted to characterise the influence of canopy position. Higher sucrose, trigonelline and caffeine contents and greater aroma intensity were found in coffee beans from the lower canopy. According to transcriptome analysis, this increase resulted from increased expression of genes likely to be associated with these bean components and slower but longer maturation in coffee beans from the lower canopy. The slower and longer maturation was likely to be related with differential expression of genes associated with hormone inhibition and slower loss of photosynthesis. This study greatly improved understanding of molecular and genetic basis of coffee quality and canopy influences. Candidate genes from this study are potential for future breeding for high quality coffee.
