The crystal structure of hypothetical protein MTH1491 from Methanobacterium thermoautotrophicum

Abstract: As part of our structural proteomics initiative, we have determined the crystal structure of MTH1491, a previously uncharacterized hypothetical protein from Methanobacterium thermoautotrophicum. MTH1491 is one of numerous structural genomics targets selected in a genome-wide survey of uncharacterized proteins. It belongs to a family of proteins whose biological function is not known. The crystal structure of MTH1491, the first structure for this family of proteins, consists of an overall five-stranded parallel… Show more

“…However, in DsrE only the first of these two cysteine residues, Cys78, is conserved. MTH1491 from the archaeon Methanobacterium thermoautotrophicum is another protein related to DsrE that has been crystallized (11). This protein forms a homotrimer.…”

“…Only a surface-exposed loop centered on residue E40 and immediately following helix ␣2, shows increased mobility. Additionally, the Cterminal pentapeptide segment shows increased mobility, as indicated by reduced 15 N{ 1 H}-NOE values (data not shown).…”

“…Heteronuclear 15 N relaxation data show that TM0979 is a rigid protein with intermolecular fluctuations limited to the sub-ns time scale. Only a surface-exposed loop centered on residue E40 and immediately following helix ␣2, shows increased mobility.…”
