The Crosstalk Between Tumor-Associated Macrophages (TAMs) and Tumor Cells and the Corresponding Targeted Therapy

Abstract: Tumor microenvironment (TME) is composed of tumor cells and surrounding non-tumor stromal cells, mainly including tumor associated macrophages (TAMs), endothelial cells, and carcinoma-associated fibroblasts (CAFs). The TAMs are the major components of non-tumor stromal cells, and play an important role in promoting the occurrence and development of tumors. Macrophages originate from bone marrow hematopoietic stem cells and embryonic yolk sacs. There is close crosstalk between TAMs and tumor cells. With the occ… Show more

“…TAMs are important players in almost every step of metastasis formation [54]. Activation of Toll-like receptor 4 (TLR-4) on the surface of M2-like macrophages increases the level of IL-10, which promotes the EMT program, which plays an important role in the first steps of metastases [55]. EMT can also be induced by proinflammatory cytokines (IL-6, IL-1β, TNFα) [56] and TGFβ [57] released by TAMs (Figure 2).…”

“…A specific example of such an influence exists in terms of CAFs, which are able to affect monocyte recruitment and polarization. Indeed, CAFs and TAMs are the main components of stromal cells (240)(241)(242)(243). Tumor associated macrophages display high level of plasticity.…”

“…Based on the gene expression profiling analysis, high-risk neuroblastoma tissues express relatively high levels of CCL2, CXCL12, and TGFB1, which could influence the recruitment and polarization of myeloid cells (58)(59)(60)(61)(62)(63)(64)(65). We therefore addressed whether the various quantities of M2 TAMs and M-MDSCs linked to the variation in the cytokine/chemokine levels in the TME.…”
