The Correlation Between the Expression of Differentiation Markers in Rat Small Intestinal Mucosa and the Transcript Levels of Schlafen 3

Abstract: IMPORTANCE The normal absorptive function and structural maintenance of the intestinal mucosa depend on a constant process of proliferation of enterocytic stem cells followed by progressive differentiation toward a mature phenotype. The mechanisms that govern enterocytic differentiation in the mucosa of the small intestine are poorly understood. OBJECTIVE To determine whether schlafen 3 (but not other schlafen proteins) act in vivo and whether its effects are limited to the small intestine. We have previousl… Show more

“…SLFN12 also inhibits LNCaP proliferation. Other Schlafen proteins have been studied in rodents (31, 32), where some tissue-specific effects on differentiation and proliferation have previously been ascribed to Slfn1 (proliferation in T-lymphocytes (33)), Slfn2 (osteoclast differentiation (13)), and Slfn3 (enterocyte differentiation (15)). Slfn4 is increased during macrophage activation by LPS, but suppressed during differentiation induced by colony-stimulating factor (CSF-1) (14).…”

“…However, a number of reference proteins for the Western blotting technique exists. When using villin, for example, colonic P-gp expression was lower than small intestinal P-gp in male rats [46]. Therefore, the lower expression of reference proteins in the colon may lead to an inflated higher calculation for the relative P-gp expression in Western blot experiments.…”

Additionally, we also looked at the brush border enzymes alkaline phosphatase (ALPI) and lactase (LCT), as well as markers of secretory cell types, including enteroendocrine (gastric inhibitory peptide (GIP) and chromogranin A (CHGA)), goblet (mucin 2 (MUC2)) and Paneth cells (lysozyme (LYSO)).…”
