The contribution of the Tie2+ lineage to primitive and definitive hematopoietic cells

Abstract: The regulatory elements of the Tie2/Tek promoter are commonly used in mouse models to direct transgene expression to endothelial cells. Tie2 is also expressed in hematopoietic cells, although this has not been fully characterized. We determine the lineages of adult hematopoietic cells derived from Tie2 expressing populations using Tie2-Cre;Rosa26R-EYFP mice. In Tie2-Cre;Rosa26R-EYFP mice, analysis of bone marrow cells showed Cre-mediated recombination in 85% of the population. In adult bone marrow and spleen, … Show more

“…(28)(29)(30) We therefore examined whether the reduced TRAP activity in Tie2-Dkk1 TG mice is associated with inhibition of osteoclast differentiation from progenitor cells. To accomplish this, we first examined in vitro differentiation of osteoclast.…”

“…The Tie2 promoter and enhancer are commonly used in mouse models to direct transgene expression to endothelial cells. (28,31,32) Indeed, we have previously demonstrated Tie2-driven overexpression of Dkk in endothelial cells. (17) Although Tie2 is expressed in all endothelial cells, (27) there is also evidence that it is expressed in various hematopoietic cells and adult bone marrow.…”

“…47 The Tie2 promoter is active in endothelial and hematopoietic lineages from E7.5 onwards, 48 and almost all primitive hematopoietic cells can be traced to Tie2 þ cells. N1ICD and Tie2 have a similar expression pattern in YS blood islands.…”

“…17 More importantly, Tie1 is expressed by a small proportion (≈20%) of BMD hematopoietic cell lineages 21 (as opposed to other constitutive endothelial-specific Cre models, such as Tie2Cre, which is reportedly expressed in ≈80%). 28 We examined β3-integrin expression in BMDCs from both Cre models. Although we saw no changes in β3-integrin in BMDCs of OHT-treated Pdgfb-iCreER T2 -positive animals, Tie1Cre-positive animals showed a loss of β3-integrin expression in CD45 + cells (CD45 is a PAN marker for hematopoietic cells, including hematopoietic progenitor/ stem cells but excluding erythrocytes and platelets; Online Figure IIB).…”
