The continuum of monocyte phenotypes: Experimental evidence and prognostic utility in assessing cardiovascular risk

Abstract: The monocyte-macrophage cell lineage represents a major player in innate immunity, and is involved in many physiologic and pathologic conditions. Particularly, monocyte-macrophages play a very important role in atherosclerosis and cardiovascular disease. Monocyte heterogeneity is well recognized but the biologic and clinical meaning of the various monocyte subtypes is not entirely understood. Traditionally, monocytes can be divided in classical, intermediate, and nonclassical based on expression of the surface… Show more

“…Results from manual analysis of flow cytometric data were further validated by means of an unsupervised clustering algorithm (CITRUS, as implemented in the cloud-based Cytobank software, Cytobank Inc. USA). This approach accounts for the continuous nature of immune subsets (e.g., monocyte subpopulations) and has been shown to better reflect pathophysiologic conditions than gating-based, threshold-driven manual analysis of distinct subtypes ( 23 ). Briefly, CITRUS performs hierarchical clustering of cellular populations based on phenotypical similarity and automatically identifies stratifying features between groups ( 24 ).…”

“…1B "nonclassical" CD14 + /lowCD16 + macrophages in the sputum, 31 which have been shown to have distinct functions. 32,33 Monocytes and macrophages from sputum were predominantly CD16 + (nonclassical; Fig. 1C) and were lower in AS patients (AS 12.2% vs. HC 25.8%).…”

“…Gating strategy based on CD14 and CD16 is the most popular classification method for the monocyte subsets among studies on coronary atherosclerosis [15]. The expression levels of CD14 and CD16 on monocyte subsets have obvious individual specificity [16]. And CD14 and CD16 expression levels will change with environmental stimuli [17,18].…”
