The Construction and Analysis of lncRNA–miRNA–mRNA Competing Endogenous RNA Network of Schwann Cells in Diabetic Peripheral Neuropathy

Abstract: Background: Diabetes mellitus is a worldwide disease with high incidence. Diabetic peripheral neuropathy (DPN) is one of the most common but often ignored complications of diabetes mellitus that cause numbness and pain, even paralysis. Recent studies demonstrate that Schwann cells (SCs) in the peripheral nervous system play an essential role in the pathogenesis of DPN. Furthermore, various transcriptome analyses constructed by RNA-seq or microarray have provided a comprehensive understanding of molecular mecha… Show more

“…Previous studies have reported that the ceRNA mechanism can be used for the analysis of multiple diseases ( 36 , 57 ). Qu et al ( 36 ) constructed a miRNA-lncRNA-mRNA ceRNA regulatory network on the basis of DEGs and lncRNAs in patients with IDD.…”

“…Qu et al ( 36 ) constructed a miRNA-lncRNA-mRNA ceRNA regulatory network on the basis of DEGs and lncRNAs in patients with IDD. In addition, Wang et al ( 57 ) identified a novel ceRNA network that provides a novel insight for investigating the underlying molecular mechanism of diabetic peripheral neuropathy. In the present study, ITGB8 was inhibited by increased expression of MREs resulting from decreased expression of lncRNAs in the ceRNA network.…”

“…Although the incidence of TN was higher in female rats than male, we only used male SD rats to establish the TN animal model, to avoid the effect of estrogen on experimental results. We only focused on transcrip-tome level changes; however, noncoding RNAs have been found to play an important role, which were not involved in our experiment (31,32). Besides the previously mentioned, we also did not identify the DEGs at the protein level.…”

“…Wang, C. et al employed whole-transcriptome sequencing technology to systematically analyze the differential expression of lncRNAs, mRNAs, and miRNAs in Schwann cells (SCs) of DPN and control rats and constructed lncRNA–miRNA–mRNA competing endogenous RNA (ceRNA) network of SCs. This network has identified the inhibited relationship of lncRNA, miRNA, and mRNA and underlined that they function as key mediators in the pathophysiological process of SCs in DPN ( Wang et al, 2020 ). This ceRNA regulatory network has a particular clinical application value in DPN.…”
