The constitutive androstane receptor and pregnane X receptor in the brain

Abstract: Since their discovery, the orphan nuclear receptors constitutive androstane receptor (CAR;NR1I3) and pregnane X receptor (PXR;NR1I2) have been regarded as master regulators of drug disposition and detoxification mechanisms. They regulate the metabolism and transport of endogenous mediators and xenobiotics in organs including the liver, intestine and brain. However, with proposals of new physiological functions for NR1I3 and NR1I2, there is increasing interest in the role of these receptors in influencing brain… Show more

“…However, the control of retinoid metabolism, inflammation, iron, glucose, lipid, cholesterol, bile acid, and bilirubin homeostasis, as well as the regulation of bone mineral density, were also demonstrated [ 19 , 70 , 75 , 76 ]. In this sense, mammalian pxr transcripts have also been detected in osteoblastic and neural cells, as well as in the endothelial cells of the blood-brain barrier (BBB) [ 17 , 18 , 19 , 77 ]. In fish, a broad distribution of pxr expression in tissues/organs was reported in different species [ 20 , 37 , 78 , 79 ].…”

“…Available data support the hypothesis that CAR functions may extend to a dialogue between multiple organs, perhaps including the central nervous system (Figure 2). The role of CAR could differ according to brain regions as suggested [134]. Furthermore, in the brain, CAR may be involved in the control of energy homeostasis through a cross-talk with the liver or the adipose tissue [13,87,133].…”

“…PXR is also observed in the THP1 cell line (acute monocytic leukemia) [35], in mouse peritoneal macrophages [39], mesenteric adipose tissue [40], and calvaria osteoblasts [41]. PXR and CAR expression profiles in placenta and brain have been reviewed recently [42,43].…”
