The composition of fluorescent pseudomonad populations associated with roots is influenced by plant and soil type

Abstract: The diversities of fluorescent pseudomonads, from two uncultivated soils and from the roots of two plant species cultivated in these two soils, were compared. The phenotypic diversity of the bacterial isolates was characterized on the basis of biochemical and physiological tests and on the basis of their ability to utilize 147 different organic compounds. The genotypic diversity of the isolates was characterized on the basis of the types of 16S genes coding for rRNA (rDNA), their repetitive extragenic palindro… Show more

“…3. Twelve ribotypes (1, 2, 3, 4, 7, 8, 9, 11, 12, 13, 18 and 20) were previously described by Laguerre et al [31] and 6 (C8, D3, C9, C5, C1, D5) by Latour et al [25]. According to the Jaccard pairwise similarity matrix, the level of similarity between the different ribotypes ranged from 0.17 to 0.97 indicating a high polymorphism of the 16S rRNA genes.…”

“…Numbers designate the hrcRST types, and letters designate the patterns obtained with the restriction enzymes AluI, AvaII, DdeI, HinfI, MspI, PstI, RsaI, and SphI, respectively. Numbers designate the 16S rDNA types as described by Laguerre et al[31], numbers following ''C'' and ''D'' designate 16S rDNA types as described by Latour et al[25], numbers following ''N'' designate new 16S rDNA types described in this study, and letters designate the patterns obtained with the restriction enzymes AluI, DdeI, HaeIII, MseI, MspI, NdeII, RsaI, and TaqI, respectively.S. “…The experiments were performed in the calcic silt^clay soil from Cha ªteaurenard (France). This soil is known for its natural suppressiveness to fusarium wilts and its main characteristics have previously been described [30]. The soil was air-dried and sieved ( 6 2 mm).…”

“…Both phenotypic tests (e.g. carbon source utilization, antibiotic resistance or fatty acid methyl ester pro¢les [29,31,32]) and genetic ¢ngerprinting techniques such as restriction analysis of ampli¢ed ribosomal DNA (ARDRA) [30,33] can be used. Other methods describe the overall function or composition of a community.…”
