The Complex Story of Plant Cyclic Nucleotide-Gated Channels

Abstract: Plant cyclic nucleotide-gated channels (CNGCs) are tetrameric cation channels which may be activated by the cyclic nucleotides (cNMPs) adenosine 3′,5′-cyclic monophosphate (cAMP) and guanosine 3′,5′-cyclic monophosphate (cGMP). The genome of Arabidopsis thaliana encodes 20 CNGC subunits associated with aspects of development, stress response and immunity. Recently, it has been demonstrated that CNGC subunits form heterotetrameric complexes which behave differently from the homotetramers produced by their const… Show more

“…CNGCs can be assembled as homotetrameric or heterotetrameric complexes, allowing for the formation of a large array of sensors capable of responding to different intensities of environmental cues ( Clough et al, 2000 ; Tan et al, 2020 ). The cytosolic C-terminus harbors a cyclic nucleotide-binding domain (CNBD) and a calmodulin-binding domain (CaMBD), in which a calmodulin (CaM) binding isoleucine-glutamine (IQ) motif is embedded ( Kaplan et al, 2007 ; Jarratt-Barnham et al, 2021 ). A. thaliana AtCNGC2 and four acts as a heat sensor since its lack of expression leads to a hyper thermosensitive phenotype, resulting in a higher accumulation of HSPs at lower temperature.…”

“…In contrast to CaM7-arrested CNGC14, the activity of other CNGCs could be positively regulated by CaM binding (e.g. CaM1 to CNGC12) (Dietrich, Moeder, & Yoshioka, 2020;Jarratt-Barnham, Wang, Ning, & Davies, 2021). These aforementioned studies have shed light on the diverse regulatory mechanism of CNGC.…”

“…CIPK proteins are activated by calcineurin B-like proteins (CBLs) and phosphorylated loop region located in the kinase domain ( Tang et al, 2020 ), and are involved in the salinity tolerance mechanisms ( D’Angelo et al, 2006 ). The members of the CNGC gene family were reported to undergo phosphorylation and post-translational modifications ( Kakar et al, 2017 ), and the phosphorylation of CNGC was believed to be responsible for defense signaling ( Jarratt-Barnham et al, 2021 ). Therefore, we speculated that LZ04 attenuates the effects of high calcium stress on A. thaliana , and related genes may be involved in this process by the regulation of calcium-dependent phosphorylation systems.…”
