The Comparative Effects of Gene Modulators on Thyroid-Specific Genes and Radioiodine Uptake

Abstract: The aim of this study was to comparatively investigate the effects of 5-azacytidine-C (5-Aza), trichostatin-A (TSA), and all-trans retinoic acid (ATRA) on mRNA expressions of Na/I symporter (NIS), thyroglobulin (Tg), thyroid peroxidase (TPO), and thyroid stimulating hormone receptor (TSH-R), and radioiodine (RAI) uptake in cancer (B-CPAP) and normal (Nthy-ori 3-1) thyroid cell lines. Cell lines were treated with 10 ng/mL of TSA, 5 microM of 5-Aza, and 1 microM of ATRA, according to the MTT (methyl-thiazol-tetr… Show more

“…For long-term storage, cell lysates were kept at -80°C. Relative TPO gene expression in the Nthy-ori 3-1 cell line has been previously quantified by others (Tuncel et al, 2007). In our experiments, TPO activity in cell extracts, which is proportional to relative fluorescence (for Ampliflu Red) or relative luminescence (for luminol) obtained from cell lysates containing 0.1-0.2 mg/ml total protein, was consistently above the limit of quantitation (LOQ) assessed as the mean of the blank + 10 times the standard deviation.…”

“…Suppression of signaling through the MAPK pathway (by knockdown of mutant BRAF or pharmacological inhibition of MEK) restores the expression of iodide-metabolizing genes in human thyroid carcinoma cells, highlighting this pathway as a critical target in re-differentiation strategies (Liu et al 2007, Hou et al 2010. On the epigenetic front, NIS and TSHR expression has been restored in human thyroid carcinoma cell lines with demethylating agents (Venkataraman et al 1999, Xing et al 2003, Tuncel et al 2007. In poorly differentiated and undifferentiated thyroid carcinoma cells, HDACi can also reactivate the expression of NIS, TPO and thyroglobulin.…”

MIO-M1, MDA-MB-453, MCF-7 were cultured in Dulbecco’s modification of Eagle’s media (DMEM) containing glutamine, supplemented with 10% heat-inactivated fetal bovine serum (FBS), penicillin (100 IU/ml) and streptomycin (100 IU/ml).…”
