A vast number of chemicals require environmental safety assessments for market authorisation. To ensure acceptable water quality, effluents and natural waters are monitored for their potential harmful effects. Tests for market authorisation and environmental monitoring usually involve the use of large numbers of organisms and, for ethical, cost and logistic reasons, there is a drive to develop alternative methods that can predict toxicity to fish without the need to expose any animals. There is therefore a great interest in the potential to use cultured fish cells in chemical toxicity testing. This review summarises the advances made in the area and focuses in particular on a system of cultured fish gill cells grown into an epithelium that permits direct treatment with water samples. KEY WORDS: FIGCS, Biomonitoring, Environmental risk assessment, Fish, In vitro, Toxicology IntroductionThe Industrial Revolution caused a rapid rise in the use of raw materials and urbanisation as the populace moved to the cities for employment. Since this time, there has been a continuous increase in living standards that to a large part has been fuelled by innovations within the chemical and pharmaceutical industry. Life expectancy has increased as a result of great advances in medical practices and effective drugs against many fatal diseases. The increase in life expectancy has seen the population of the world grow, reaching 7 billion in 2012, and to feed this population there have been great advances in agricultural productivity partly via the development of pesticides and nitrate/phosphate-based fertilisers. These activities have altered the geochemical cycling of elements, increasing or decreasing concentrations in earth system compartments and increasing global distribution (Doney, 2010). Anthropogenic activities have left a cumulative and lasting impression on the biosphere -so much so that geologists have termed the current epoch the anthropocene (Zalasiewicz et al., 2010).The increase in agricultural and industrial production and consumption of raw materials has resulted in the creation of vast amounts of waste that enters the aquatic ecosystem. An acknowledgement of the decline in environmental quality due to contaminants has led to the development of environmental quality standards (EQS) in many countries, and to assess whether these standards are being adhered to, many jurisdictions also have a programme of waste water effluent testing (WET) and/or biomonitoring. The EQS are derived from toxicity tests that use numerous organisms per compound, and in order to set standards several species are tested. In the USA ~3 million fish are used in WET procedures (see Tanneberger et al., 2013). An expansion of anthropogenic activity around Lagos lagoon, Nigeria, has raised concerns over increasing contaminants entering the lagoon’s ecosystem. The embryotoxicity, teratogenicity and genotoxicity of sediment organic extracts from four sampling zones around Lagos lagoon, Ilaje, Iddo, Atlas Cove and Apapa, as well as the dominant polycyclic aromatic hydrocarbons (PAHs) identified in water measured during the wet season (naphthalene, phenanthrene, pyrene, benzo[a]pyrene and a mixture of these), were assessed with Danio rerio embryos. Embryos were exposed to varying concentrations of toxicants from 0–72 h post-fertilization (hpf). Embryotoxicity at 72 hpf showed a dose-dependent increase in mortality upon exposure to extracts from all zones, except Atlas Cove. Similarly, higher levels of teratogenic effects, such as increased oedema, and haemorrhage and developmental abnormalities resulted from exposure to extracts from Ilaje, Iddo and Apapa zones. Treatment with single PAHs revealed that significant levels of detrimental effects were obtained only for phenanthrene. The modified comet assay revealed that the oxidative damage to DNA was generally low (<12 %) overall for all sediment extracts, but was significantly elevated with Ilaje and Iddo sediment extracts when compared with solvent controls. Oxidative damage was observed with the single PAHs, phenanthrene and benzo[a]pyrene, as well as with the PAH mixture. This study highlights that Lagos lagoon sediment extracts have teratogenic, embryotoxic and genotoxic properties, which are likely due to the high molecular weight PAHs present in the extracts, some of which are known or are suspected human carcinogens.

Industrial activity generates harmful substances which can travel via aerial or water currents thousands of kilometers away from the place they were used impacting the local biota where they deposit. The presence of harmful anthropogenic substances in the Antarctic is particularly surprising and striking due to its remoteness and the apparent geophysical isolation developed with the flows of the Antarctic Circumpolar current and the ring of westerly winds surrounding the continent. However, long-range atmospheric transport (LRAT) of pollutants has been detected in the Antarctic since the 70’s along the Antarctic trophic food web from phytoplankton to birds. Still, no information exists on the presence of cytotoxic compounds in marine sediments neither at basin scales (thousands of kilometers) nor in water depths (hundreds of meters) beyond shallow coastal areas near research stations. Our results showed for the first time that there is cytotoxic activity in marine sediment extracts from water depths >1000 m and along thousands of kilometers of Antarctic continental shelf, in some cases comparable to that observed in Mediterranean areas. Ongoing anthropogenic pressure appears as a serious threat to the sessile benthic communities, which have evolved in near isolation for millions of years in these environments.
