The co‑treatment of metformin with flavone synergistically induces apoptosis through inhibition of PI3K/AKT pathway in breast cancer cells

Abstract: Abstract. Metformin, a widely used antidiabetic drug, exhibits anticancer effects which are mediated by the phosphatidylinositol 3-kinase (PI3K)/serine/threonine kinase (AKT) signaling pathway. However, its use in anticancer therapy combined with other natural products remains unclear. Flavone as the core structure of flavonoids has been demonstrated to induce cell apoptosis without causing serious side effect. Murine double minute X (MDMX) inhibits tumor suppressor gene p53 whose function is associated with t… Show more

“…Therefore, the reduction of the concentration and toxicity of ADM under the premise of ensuring the therapeutic effect is a key problem in the treatment of breast cancer. Recently, many kinds of combined drugs are commonly used in the treatment of tumors, and good curative effects have been achieved (Zheng et al, 2018). Treatment with a combination of ADM and Taxol (paclitaxel) is a current clinical recommendation (Ardavanis et al, 2008), but its application is hampered by the collection of the raw material for paclitaxel, whereby rare plant resources are destroyed (Gelderblom et al, 2001).…”

“…As a transcription factor, p53 targets multiple elements involved in the apoptotic pathway, including p53-regulated apoptosis-inducing protein 1 (p53AIP1) and pro-apoptotic Bcl-2 family members (Chipuk et al, 2004). Additionally, p53 is upregulated by the phosphorylation of p38 MAPK (Dewanjee et al, 2017), elevated p53 is related to the mediation of apoptosis (Zheng et al, 2018). The activated JNK can regulate a variety of transcription factors and mitochondrial proteins like Bcl-2 and Bcl-xL (Radogna et al, 2015).…”
