Freeze-dried black raspberries (BRB), their component anthocyanins (AC), and a metabolite of BRB ACs, protocatechuic acid (PCA), inhibit the development of esophageal cancer in rats induced by the carcinogen, N-nitrosomethylbenzylamine (NMBA). All three components reduce inflammation in the esophagus and in plasma. The present study determined the relation of changes in inflammatory markers to infiltration of innate immune cells into NMBA-treated esophagus. Rats were injected with NMBA (0.35 mg/kg) for 5 weeks while on control diet. Following NMBA treatment, rats were fed diets containing 6.1% BRB powder, an AC-rich fraction of BRBs (3.8 mmol/g), or 500 ppm PCA. At weeks 15, 25, and 35, inflammatory biomarker expression in the plasma and esophagus was quantified, and infiltration of immune cells in the esophagus was examined. At all three time points, BRB, AC, and PCA similarly affected cytokine production in the esophagus and plasma of NMBA-treated rats, relative to the NMBA-only control. These included decreased expression of the proinflammatory cytokine IL1b and increased expression of the anti-inflammatory cytokine IL10. Moreover, all three diets also increased the expression of IL12, a cytokine that activates both cytolytic natural killer and CD8 þ T cells. In addition, the three diets also decreased infiltration of both macrophages and neutrophils into the esophagus. Overall, our results suggest that another mechanism by which BRBs, ACs, and PCA inhibit NMBA-induced esophageal tumorigenesis is by altering cytokine expression and innate immune cell trafficking into tumor tissues.

Objectives: The most active organs of the reticuloendothelial system (RES) are the liver, spleen and bone marrow, having immune mechanisms against malignancy including neutrophils and platelets. RES may be imaged by different modalities, like PET scan. Neutrophil lymphocyte ratio (NLR) and platelet lymphocyte ratio (PLR) have gained importance as proinflammatory markers in cancer, e.g. lung cancer. The aim of this study was to investigate the relationship between PET parameters of pulmonary mass or RES, and hematological parameters, and to evaluate the role of these factors in differentiating the pathological character of the mass. Methods:A retrospective analysis of the data of 131 patients, retrieved from the department archives, with pulmonary mass limited to mediastinum was made. Patients were grouped according to pathological results: benign mass (n=46), squamous cell carcinoma (n=38), and non-squamous cancer of lung (n=47). All patients underwent PET/CT scanning and images were analyzed retrospectively. Maximum and mean SUV were calculated from primary lesions and RES. NLR and PLR were calculated from CBC.Results: SUV max and SUV mean of RES organs were similar for both groups with benign and malignant pulmonary masses, and among the subgroups. SUV max ratios of pulmonary mass were significantly different between the groups (the highest value in the squamous cell carcinoma and the lowest in the benign groups). No significant difference was determined between the subgroups for NLR and PLR. NLR was significantly correlated with SUV max ratios of spleen and the mass, and SUV mean ratios of spleen and bone marrow. PLR was significantly correlated with SUV max ratios of spleen, bone marrow, the mass and SUV mean ratios of spleen, bone marrow. Conclusion:SUV of RES and primary mass were correlated with NLR and PLR, indicators of systemic inflammation. The associations between NLR and PLR, and SUV should be clearly defined by further investigations.

Comparative prognostic value of different preoperative complete blood count cell ratios in patients with oral cavity cancer treated with surgery and postoperative radiotherapy

Background We sought to compare the prognostic significance of different preoperative complete blood count cell ratios in patients with oral cavity squamous cell carcinoma (OSCC) treated with surgery and postoperative radiotherapy (PORT). Methods We retrospectively reviewed the clinical records of 890 patients with OSCC who were treated with surgery and PORT. The following preoperative complete blood count cell ratios were collected: neutrophil‐to‐lymphocyte ratio (NLR), platelet‐to‐lymphocyte ratio (PLR), and lymphocyte‐to‐monocyte ratio (LMR). Overall survival (OS), local control, regional control, and distant control (DC) served as the main outcomes of interest. Results The results of multivariate analysis in the entire study cohort revealed that a low NLR was the only independently favorable marker of both OS (adjusted hazard ratio [HR]: 0.794, 95% confidence interval (CI): 0.656–0.961, bootstrap p = 0.028) and DC (adjusted HR: 0.659, 95% CI: 0.478–0.909, bootstrap p = 0.015). Both LMR and PLR were not retained in the model as independent predictors. Subgroup analyses in high‐risk patients (i.e., those bearing T4 disease, N3 disease, or poor differentiation) revealed that a high NLR was a significant adverse risk factor for both OS and DC (all p < 0.03)—with a borderline significance being evident for DC in patients with T4 disease (p = 0.058). Conclusions A high pretreatment NLR was an independent unfavorable risk factor for both OS and DC in patients with OSCC who underwent surgery and PORT. No other preoperative complete blood count parameters and cell ratios were found to have prognostic significance.
