The Class III peroxidase gene OsPrx30, transcriptionally modulated by the AT‐hook protein OsATH1, mediates rice bacterial blight‐induced ROS accumulation

Abstract: Class III peroxidases (CIII Prxs) play critical roles in plant immunity by scavenging reactive oxygen species (ROS). However, the functions of CIII Prxs in rice (Oryza sativa L.) immunity are largely unexplored. Here, we report a Prx precursor, OsPrx30, that is responsive to the bacterial blight Xanthomonas oryzae pv. oryzae (Xoo). OsPrx30 was primarily expressed in rice roots, leaves, and stems,

“…The AT‐hook facilitates binding specificity to AT‐rich DNA sequences at nuclear matrix attachment regions (MARs), whereas DUF296 enables protein‐protein interactions. The AHL protein has diverse roles in the plant developmental process, including hypocotyl elongation, gibberellin biosynthesis, pollen nexine formation and flower development (Liu et al ., 2021a). AT‐rich sequences are generally found in flanking regions such as the transcriptional units or regions close to cis ‐regulatory elements.…”

“…Several TFs bind to PRX promoters to activate or repress their expression and modulate POD activity, which is involved in lignified tissue formation, seed longevity, disease resistance and abiotic stress tolerance in plants (Huang et al, 2013;Liu et al, 2015Liu et al, , 2021Cosio et al, 2017;Renard et al, 2020). However, the TFs involved in regulating PRX-mediated H 2 O 2 scavenging under multiple abiotic stress conditions are still poorly understood.…”

“…Subcellular localization markers fused to mCherry include the nuclear marker NLS-mCherry, the endoplasmic reticulum (ER) marker HDEL-mCherry, the Golgi marker Man49-mCherry, and the membrane marker PIP2A-mCherry (Nelson et al, 2007). Rice protoplasts were prepared as previously described (Liu et al, 2021). Protoplasts were resuspended in WI solution (4 mM MES, 0.5 M mannitol, 20 mM KCl, pH 5.8) for 16-22 h in the dark after transient transfection.…”

“…Only very limited information is available about the regulation of secretory peroxidases. Their transcriptional control is mediated by AT-hook protein OsATH1 in rice ( Liu et al, 2021 ) or by AGAMOUS-LIKE15 (AGL15) in Arabidopsis ( Cosio et al, 2017 ). The mechanism of MAPK-dependent regulation of peroxidases is not resolved yet.…”
