In Mexico, Diabetes mellitus (DM) is a serious health problem, and although the current pharmacological treatments for DM such as insulin and oral hypoglycemics are available, the Mexican population continues to use medicinal plants in the treatment of DM. The antidiabetic properties of the plant species that belong to the Cucurbitaceae family has already been recognized worldwide. Since Mexico is one of the most important centers of diversity of Cucurbitaceae, the present work contributes to the review of the most used species of Cucurbitaceae in the treatment of DM in Mexico. The reviewed species (Cucurbita ficifolia, C. maxima, C. moschata, C. pepo, Ibervillea sonorae, Sechium edule, Citrullus lanatus, Cucumis melo, and C. sativus) revealed that the antidiabetic effects exerted are effective in a number of mechanisms involved in the complex pathogenesis of DM: hypoglycemic, antioxidant, anti-inflammatory, anti-obesity, protective effects on diverse organs and cells, as well as in the control of dyslipidemias; furthermore, the select species of the Cucurbitaceae family could also be essential components of diets for the control of DM in patients with the disease. Thus, the Cucurbitaceae species selected in the present work represent a source of antidiabetic agents that perhaps establish the bases for novel clinical treatments.

The present study reports the structural characteristics of three polysaccharide fractions (SPS-F1, SPS-F2 and SPS-F3) isolated and purified from squash. SPS-F1(Mw=12.30 kDa) and SPS-F2 (Mw=19.40 kDa) were likely to contain HG and RG-I domain of pectic polysaccharide, respectively. SPS-F2 (Mw=270.4 kDa) was mainly composed of rhamnose, galactose and arabinose. The treatment with SPS decreased body weight gain, glucose and TG levels in T2DM rats. Twenty-five differential metabolites were identified based on urinary metabolomics analysis, which are crucial to the anti-diabetic effect of SPS. The regulation of Nicotinamide N-oxide, Histamine, cis-Aconitate, Citrate, L-Malic acid, 3-(3-Hydroxyphenyl) propanoic acid and N-Acetyl-L-aspartic acid was mainly associated with energy metabolism, gut microbiota and inflammation. Study of surface plasmon resonance revealed the binding kinetics with galectin-3 (Gal-3) and fibroblast growth factor 2 (FGF2). The K D value of SPS-F2 and SPS-F3 to Gal-3 are 4.97 × 10 -3 M and 1.48 × 10 -3 M, indicating a weak binding affinity. All three fractions showed moderate binding to FGF2 and the affinity was SPS-F3 > SPS-F2 > SPS-F1. Thus, the metabolomics and SPR approach were proved to be a promising tool in exploring the anti-diabetes effects of SPS and provided a deep understanding of the mechanisms.

Structure of pumpkin pectin and its effect on its technological properties

The aim of this work is to investigate the structural features, physicochemical, rheological, and emulsifying properties of pectin extracted from pumpkin “Cucurbita maxima” pulp and peel. After the peel and pulp of fresh pumpkin were solubilized in water at high temperature (80°C) and clarification, the obtained filtrate was treated with aluminum sulphate, which precipitates a fibrous coagulum. The results found that the major functional groups of pectin isolates identified by Fourier transform infrared are: –OH, CH2, C═C, –C–O–C–, and CH3. The results obtained using nuclear magnetic resonance methods suggest that these polysaccharides consist of two domains: the first one is homogalacturonan and the second is formed by rhamnogalacturonan. Mineral and metal ions on pectin surface detected using scanning electron microscopy coupled to energy dispersive X-ray analysis were different pulp and peel pectin. Rheological behavior of these biopolymers is suitably represented by the Herschel–Bulkley and power law models. The extracted pectin has a viscosity spectrum expressed by the following properties: G′, G′′, G*, and ∣ η ̇ ∣ ( ω ) | \dot{\eta }| (\omega ) which, respectively, have the following values: 0.28–57.63, 0.15–34.38, 0.39–67.12, and 1.82–831.08 Pa s. These macromolecules have a negative charge on their surface. Rheological properties and emulsifying activity are significantly influenced by the structural composition and physicochemical properties.
