Isoflavone mono-O-glycosides were investigated by electrospray ionization tandem mass spectrometry with a quadrupole linear ion trap mass spectrometer in negative ion mode. Isoflavonoids having different positions of glycosylation or methylation were differentiated according to the relative abundances of Y(0)(-) and [Y(0)-H](-•) ions generated from the [M-H](-) ion. It is found that the site of glycosyl or methyl group significantly affects relative abundances of the Y(0)(-) and [Y(0)-H](-•) ions. In addition, the characteristic ion [Y(0)-2H](-) was observed in the product ion spectrum of genistein 7-O-β-D-glucoside and was also detected, together with the [Y(0)-CH(3)](-•) and [Y(0)-H-CH(3)](-) ions in the product ion spectra of glycitin and 6-methoxy genistein 7-O-β-D-glucoside. The structures of isoflavonoids can be characterized and identified according to the formation of these diagnostic ions. The results obtained from this investigation can promote the rapid identification of isoflavonoids in crude plant extracts.

Context: Three clover [Trifolium L. (Leguminosae)] species were selected on the basis of data from traditional medicine, phytochemical profiles, and agricultural significance. Objective: The in vitro evaluations of free radical scavenging properties, ferric reducing abilities, and antioxidant effects of extracts from T. pratense L. (crude extract and phenolic fraction), T. pallidum L., and T. scabrum L. (phenolic fractions) were performed. Materials and methods: Activities of the Trifolium extracts were determined at their final concentrations of 1.5-50 mg/ml. Free radical scavenging properties of methanol extract solutions were estimated by the reduction of DPPH and ABTS radicals. Measurements of the total antioxidant capacity (TAC) were carried out to assess the antioxidant activities of the extracts in human blood plasma under conditions of oxidative stress, induced by 200 mM peroxynitrite. Results: The phenolic fraction of T. pratense displayed the strongest ABTS and DPPH radical scavenging effects (EC 50 value of 21.69 and 12.27 mg/ml, respectively). The EC 50 value for T. pallidum extract attained 29.77 and 30.06 mg/ml. The two remaining extracts were less potent scavengers (EC 50 value higher than 50 mg/ml). Similar differences were obtained during evaluation of the ferric reducing abilities. Analysis of antioxidant properties of the extracts in blood plasma did not provide such evident differences in their actions, however, it indicated that the T. pratense phenolic fraction displayed the strongest effect. Conclusions: The examined Trifolium extracts partly protected blood plasma and enhanced its non-enzymatic antioxidant defense against harmful action of peroxynitrite in vitro.

Caracterização das plantas medicinais e medicamentos fitoterápicos para tratamento da osteoporose utilizados no Brasil. Arq. Cienc. Saúde UNIPAR, Umuarama, v. 20, n. 3, p, 183-188, set./dez. 2016. RESUMO:A osteoporose é caracterizada por desordem do sistema esquelético caracterizada por baixa massa óssea e deteriorização da microarquitetura do tecido ósseo levando a fragilidade óssea com aumento da suscetibilidade a fraturas. Considerada a doença osteometabólica mais comum, de distribuição global, pode comprometer ambos os sexos, diferentes idades ou raças. As consequências relacionadas a essas doenças são as fraturas vertebrais verificadas em aproximadamente 20% das mulheres acima de 65 anos em pós-menopausa. Além disso, a consequência dessa fragilidade óssea pode levar a consequências graves como dor, deformidades, sequelas, restrições motoras e morte precoce, portanto, associada com alta morbi/mortalidade. Desta forma, sabendo que as medicações utilizadas para o tratamento da osteoporose não são isentas de risco (por exemplo, aumento do risco de tromboses, câncer de mama/ ovário/ endométrio, lesões musculares, entre outras), foi realizado uma revisão bibliográfica com o intuito de avaliar se existem plantas medicinais que possam ter efeitos ósseo protetores com menor número de efeitos adversos em relação aos fármacos sintéticos. Em síntese, verificou-se que os fármacos sintéticos podem acarretar prejuízos à saúde do paciente a longo prazo e que as plantas medicinais ou fitoterápicos utilizadas no Brasil apresentam estudos científicos com menores efeitos colaterais e, portanto, são uma alternativa viável para o tratamento e prevenção da osteoporose, visto que a Politica Nacional de Práticas Integrativas e Complementares na Atenção Básica sustenta esta tendência mediante subsídios como a Farmacopeia Brasileira e o Memento Fitoterápico. Considered the most common osteometabolic disease worldwide, it affects both sexes, different ages and races. The consequences of these diseases are vertebral fractures in approximately 20% of postmenopausal women over 65 years of age. In addition, the consequence of this bone fragility may lead to serious consequences, such as pain, deformities, sequelae, motor restrictions and early death, being therefore associated with high morbidity/mortality.
