The cDNA Sequencing Project

Abstract: The Dictyostelium discoideum cDNA sequencing project started in 1995, preceding the genome sequencing project. Altogether, 14 cDNA libraries, including full-length ones, were constructed from five different stages of growth and asexual and sexual development, from which nearly 100,000 randomly chosen clones were sequenced to yield over 150,000 expressed sequence tags (ESTs). The data have been publicized online to facilitate clone distribution and collaboration using the whole clone set for microarray analyses… Show more

“…The authors thank S. Mitchell for Dictyostelium SBDS protein purification, the Japanese cDNA project 36 for providing clone VHA344, dictyBase for expression plasmids, N. Perrimon for plasmid pS5⌬H-G4MINT, and J. Rommens for lymphoblastoid cell lines. They thank F. Begum, S. Peak-Chew, and E. Stephens for mass spectrometry, as well as members of A.J.W.…”

“…The genome sequence is maintained and annotated at dictyBase (http://dictybase.org/), which also runs the Dictyostelium Stock Center. Also, a largescale EST project has generated cDNAs representing 55% of the estimated genes, and many cDNAs are available as full-length inserts (Urushihara et al 2006).…”
