The Carbohydrate-linked Phosphorylcholine of the Parasitic Nematode Product ES-62 Modulates Complement Activation

Abstract: Parasitic nematodes manufacture various carbohydrate-linked phosphorylcholine (PCh)-containing molecules, including ES-62, a protein with an N-linked glycan terminally substituted with PCh. The PCh component is biologically important because it is required for immunomodulatory effects. We showed that most ES-62 was bound to a single protein, C-reactive protein (CRP), in normal human serum, displaying a calcium-dependent, high-avidity interaction and ability to form large complexes. Unexpectedly, CRP binding to… Show more

“…In plasma, CRP exists as a cyclic, noncovalent pentamer of 125 kDa composed of five identical subunits (pCRP), and which is stabilized by numerous electrostatic and Van-der-Waals interactions ( 54 , 55 ). Native pCRP binds in a Ca 2+ -dependent manner to phosphocholine (PCh)-containing ligands such as pneumococcal cell wall C-polysaccharide, but also to the surface of necrotic cells and parasites ( 55 58 ). Oxidative stress, low pH, and bioactive lipids from activated or damaged cells can dissociate the CRP pentamer into its 23-kDa subunits ( 59 62 ).…”

“…With few exceptions [33,34], the identification of molecules from parasitic helminths has proved challenging. Characterization of the nature of the HdE revealed that the component that blocked neutrophil chemotaxis was a heat-and trypsin-sensitive glycan (SMP-sensitive), most likely a proteoglycan that was resistant to acidification.…”

“…The flexible nature of PC attached to CRP leads to the uncoupling of C1s and C4b, resulting in a failure of C2 cleavage. Moreover, ES-62 may be associated with the degradation of early complement components such as C4, which can also help suppress complement activation ( Ahmed et al, 2016 ).…”
