The breadth of FGF21's metabolic actions are governed by FGFR1 in adipose tissue

Abstract: FGF21 is a multifunctional metabolic regulator. The co-factor βKlotho (KLB) allows FGF21 to signal via FGF receptors. Given the widespread nature of FGFR expression and KLB presence in several organs, it remains unclear which tissue/FGFR isoform determine FGF21 action. Here we show that deletion of FGFR1 in fat (FR1KO) leads to a complete ablation of FGF21 stimulated transcriptional activity in this tissue. Furthermore, FR1KO mice showed no FGF21-mediated lowering of plasma glucose, insulin and triglycerides, … Show more

“…Mice lacking KLB are resistant to both acute and chronic effects of FGF21. However, the acute insulin-sensitizing effects of FGF21 are also absent in mice with specific deletion of adipose KLB (32) or FGFR1 (33), consistent with the notion that direct adipose tissue activation is required for FGF21 action (34).…”

“…The beneficial effects of FGF21 on glucose metabolism and body weight have identified it as a central player in regulating metabolic processes, and adipose tissue is required for its antidiabetic actions (32,33,63). These findings show that FGF21 signaling is modulated by Rev-erb␣ in WAT, not by altering FGF21 levels but through the regulation of its co-receptor KLB in WAT but not in liver.…”

“…In contrast to FGF15/19, the glycemic effects of FGF1 and FGF21 are dependent on FGFR1 activation in adipose tissue (9,26). FGF21 also can alleviate endoplasmic reticulum stress-induced hepatic steatosis by acting as a metabolic effector of the unfolded protein response (27). Whether these effects are directly mediated through FGFR activation in the liver and whether FGF1 and FGF15/19 act through the same pathway is not known.…”

“…Fibroblast growth factor (FGF)21, a metabolic hormone predominantly produced by liver, has many functional similarities to adiponectin in regulating glucose and lipid metabolism and insulin sensitivity (33)(34)(35)(36)(37). FGF21 acts through a cell-surface receptor composed of a canonical FGF receptor (FGFR) associated with a coreceptor, b-Klotho (KLB) (36)(37)(38).…”
