Abstract. Optical techniques are one of the most promising non-invasive technologies for the diagnosis of medical conditions. This work is devoted to the current state of the optical non-invasive diagnostics (OND), especially about instrumentation as well as methodological and metrological provision for this technology. In some details Laser Doppler Flowmetry (LDF) and Fluorescence Spectroscopy (FS) diagnostics methods are considered. The main conclusion is that it is necessary to solve the problems of metrological support OND devices of these types. It is also important to develop a methodology based on diagnostic criteria obtained from a relationship between a number of OND methods and others. © 2016 Journal of Biomedical Photonics & Engineering.Keywords: optical non-invasive diagnostics, blood microcirculation, biotissue, laser Doppler flowmetry, fluorescence spectroscopy.

We report a reconstruction method to achieve high spatial resolution for hyperspectral imaging of chromophore features in skin in vivo. The method utilizes an established structure-adaptive normalized convolution algorithm to reconstruct high spatial resolution of hyperspectral images from snapshot low-resolution hyperspectral image sequences captured by a snapshot spectral camera. The reconstructed images at chromophore-sensitive wavebands are used to map the skin features of interest. We demonstrate the method experimentally by mapping the blood perfusion and melanin features (moles) on the facial skin. The method relaxes the constrains of the relatively low spatial resolution in the snapshot hyperspectral camera, making it more usable in imaging applications. The paper presents the results of in vivo experimental measurements of the endogenous fluorescence and blood perfusion in common bile duct tissues in obstructive jaundice patients during minimally invasive surgical operations. Nowadays the type of interventions become more widespread for treating hepatopancreatobiliary pathologies. An effective feedback which can be established by one or more optical diagnostic channels integrated into the standard instruments for minimally invasive manipulations would be of particular value to practical surgeons. To collect reference data, an experimental study has been conducted using a fiber optical probe with fluorescence spectroscopy and laser Doppler flowmetry channels. For every measurement, the areas of interest were different areas of common bile duct obstructed by a malignant tumor. Obtained results demonstrated significant variety registered intensities of fluorescence spectra and values of blood perfusion. Tested fiber optical endoscopic system showed sensitivity to the tissue state in the investigated area, so the two modal approach seems to be promising for further research and implementation in clinical practice.
