The Biological Effects of IL-21 Signaling on B-Cell-Mediated Responses in Organ Transplantation

Abstract: Antibody-mediated rejection has emerged as one of the major issues limiting the success of organ transplantation. It exerts a highly negative impact on graft function and outcome, and effective treatment is lacking. The triggers for antibody development, and the mechanisms leading to graft dysfunction and failure, are incompletely understood. The production of antibodies is dependent on instructions from various immunocytes including CD4 T-helper cells that secrete interleukin (IL)-21 and interact with antigen… Show more

“…It is well-known that local ectopic GCF is a key event in plasma cell differentiation, in which interlukin-6 (IL-6) and Tfh cells play a central role (12,15,16). With the stimulation of IL-6, Tfh cells could be activated and promote antibody secretion of B cells and its differentiation into plasma cells with the collaboration of BAFF, CD40L, IL-21, and IL-4 (17)(18)(19)(20)(21)(22). In patients with IgG4-RD, a significantly increased number of Tfh cells was seen in the peripheral blood, tissue, tertiary lymphoid organs, and secondary lymphoid organs (18,(23)(24)(25)(26).…”

“…(14) B-cell maturation into plasma cells within the germinal centre is facilitated by Th 2 -type cytokines IL-4 (15) and IL-6 (16,17) and T FH -derived IL-21. (18) Matured plasma cells produce antibody directed against the inciting donor antigen, and this donor-specific antibody (DSA) results in humoral rejection.…”
