The Barretos Cancer Hospital Animal Facility: Implementation and Results of a Dedicated Platform for Preclinical Oncology Models

Abstract: The Barretos Cancer Hospital Animal Facility (BCHAF) is a unique facility in Brazil exclusively dedicated to working with animal models for cancer research. In this article, we briefly present our modern facility and the main experiments performed, focusing on mutant strains of mice (PTCH-knockout and ApcMin mice), xenograft models, and patient-derived xenografts (PDXs). Our results show the progress and challenges in establishing these models and the need for having an appropriate representation of our cancer… Show more

“…The combination of TMZ with carbonic anhydrase inhibitors [52], including Indisulam [20], results in increased TMZ efficacy and indicates a promising strategy toward more effective treatments. In addition, Indisulam has already been tested in combination with another chemo-and radiotherapeutic agents in vitro, in vivo, and clinical studies, showing promising results as an adjuvant strategy [20,21,[83][84][85]. Despite the limitation of working with established cell lines and in vitro models, several studies and different research groups with the purpose of better understanding the tumor biology of pediatric glioblastoma have already used the SF188 and KNS-42 cell lines, being two reliable models of investigation of this tumor subtype.…”

“…To assess the tumorigenicity of HCB-541, subcutaneous injections of either 1 × 10 6 or 3 × 10 6 cells mixed with Matrigel® at a 1:1 (v/v) ratio were administered into the right flank of 8-week-old female NSG mice (NOD.Cg-Prkd cscid Il2rg tm1Wjl /SzJ, The Jackson Laboratory, USA), as previously described [ 14 ]. Tumor size was measured weekly with calypter, and volume was calculated using the formula: volume = length × width2/2.…”

“…Comercial human glioblastoma cells (U251 and U87), and primary glioblastoma cell line (HCB 151) derived from surgical biopsies obtained in the Neurosurgery Department of Barretos Cancer Hospital ( Martinho et al, 2013 ; Teixeira et al, 2022 ) (Sao Paulo, Brazil), acquired by the local ethics committee approval and the patient’s consent agreement, were kindly donated from Dr. Rui Reis, Barretos Cancer Hospital. Cell authentication was performed through short tandem repeat (STR) DNA typing, according to the International Reference Standard for Authentication of Human Cell Lines.…”
