The Balance of TNF Mediated Pathways Regulates Inflammatory Cell Death Signaling in Healthy and Diseased Tissues

Abstract: Tumor necrosis factor alpha (TNF; TNFα) is a critical regulator of immune responses in healthy organisms and in disease. TNF is involved in the development and proper functioning of the immune system by mediating cell survival and cell death inducing signaling. TNF stimulated signaling pathways are tightly regulated by a series of phosphorylation and ubiquitination events, which enable timely association of TNF receptors-associated intracellular signaling complexes. Disruption of these signaling events can dis… Show more

“…In addition, RIP1 restricts ZBP1‐mediated necroptosis by interfering with RHIM‐mediated interaction of ZBP1 and RIP3 (Lin et al , 2016; Newton et al , 2016b). The interplay of apoptosis and necroptosis is apparent in many inflammatory in vivo models where they are frequently concomitantly activated (Webster & Vucic, 2020). This is not surprising given that majority of signaling proteins are common to both pathways, and the balance of expression or activation of critical factors (RIP3, caspase‐8) can tip the balance in favor of apoptosis or necroptosis.…”

“…TNFα triggered pro-inflammatory gene expression through the activation of both NF-κB and MAP kinase pathways ( Kyriakis and Avruch, 2001 ; Zelová and Hošek, 2013 ; Webster and Vucic, 2020 ). Therefore, we determined the two main signalling cascades activated by TNFα after TNFAIP1 silencing.…”

“…Proper regulation of cell death is critical for the development and homeostasis in metazoans. Dysregulation of cell death pathways is associated with a number of human diseases, including cancer, tissue damage/inflammation, and neurodegeneration 1 , 2 . The best-known form of regulated cell death is apoptosis, which relies on the activation of cysteine proteases called caspases 3 .…”
