The Baculovirus p35 Protein Inhibits Fas- and Tumor Necrosis Factor-induced Apoptosis

Abstract: The baculovirus p35 gene product inhibits virally induced apoptosis, developmental cell death in Caenorhabditis elegans and Drosophila, and neuronal cell death in mammalian systems. Therefore, p35 likely inhibits a component of the death machinery that is both ubiquitous and highly conserved in evolution. We now show for the first time that p35 also inhibits Fas- and tumor necrosis factor (TNF)-induced apoptosis. Additionally, p35 blocks TNF- and Fas-induced proteolytic cleavage of the death substrate poly(ADP… Show more

“…However, this caspase inhibition only partially improved thymocyte survival. Consistent with previous studies in cell lines (42,43), p35 efficiently suppressed anti-Fas-induced cell death, in which caspase-8 is known to play a primary, death-initiating role (reviewed in Ref. 44).…”

“…The baculovirus Autographa californica p35 gene product inhibits apoptosis in insects, mammals and nematodes, suggesting that it acts at a central and evolutionarily conserved part of the apoptotic pathway [24,138,146,147]. Purified recombinant p35 inhibits the activity of purified recombinant caspases, including caspases-1, -2, -3 and -4, and maximum inhibition is achieved at equimolar concentrations of p35 and the caspase.…”

“…Baculovirus-derived p35 directly binds with activated caspases and inhibits apoptosis [36,37]. Thus, it can prevent apoptosis induced by the TNF/TNFR and FasL/ Fas systems [38].…”
