The Autophagy Regulatory Molecule CSRP3 Interacts with LC3 and Protects Against Muscular Dystrophy

Abstract: CSRP3/MLP (cysteine-rich protein 3/muscle Lim protein), a member of the cysteine-rich protein family, is a muscle-specific LIM-only factor specifically expressed in skeletal muscle. CSRP3 is critical in maintaining the structure and function of normal muscle. To investigate the mechanism of disease in CSRP3 myopathy, we performed siRNA-mediated CSRP3 knockdown in chicken primary myoblasts. CSRP3 silencing resulted in the down-regulation of the expression of myogenic genes and the up-regulation of atrophy-relat… Show more

“…Autophagy plays an important role in the process of spinal muscular atrophy and denervated muscle atrophy. Autophagy inhibition can effectively relieve muscle atrophy, and autophagy is a key target for the treatment of muscle atrophy (Piras et al, 2017;Cicardi et al, 2019;Wang et al, 2019;Cui et al, 2020). Our findings demonstrate that isoquercitrin can significantly inhibit mitophagy caused by denervation of the target muscle, accompanied by decreased expression of the autophagy-related proteins ATG7, BNIP3, and PINK1.…”

“…Mutations in these proteins have been linked to various muscle-related disorders and heart diseases 35 . CSRP3 is an autophagy-regulating protein essential for degrading muscle-related components and maintaining muscle structure and function 36 . TCAP is a giant elastic protein with kinase activity that extends half the length of a sarcomere and maintains cellular structure 37 .…”

“…Of note, MLP expression levels have been found increased in muscles from mouse models and human patients affected by different types of myopathies [ 192 , 193 , 194 ]. Recently, MLP has been also involved in promoting autophagosome formation by interacting with LC3, protecting myocytes from apoptosis [ 195 ]. MLP localizes at costameres, where it interacts with zyxin [ 196 ], β1-spectrin [ 197 ] and ILK [ 198 ].…”
