The Attenuating Effect of Low-Intensity Pulsed Ultrasound on Hypoxia-Induced Rat Chondrocyte Damage in TMJ Osteoarthritis Based on TMT Labeling Quantitative Proteomic Analysis

Abstract: Temporomandibular joint osteoarthritis (TMJOA) is a degenerative disease with a complex and multifactorial etiology. An increased intrajoint pressure or weakened penetration can exacerbate the hypoxic state of the condylar cartilage microenvironment. Our group previously simulated the hypoxic environment of TMJOA in vitro. Low-intensity pulsed ultrasound (LIPUS) stimulation attenuates chondrocyte matrix degradation via a hypoxia-inducible factor (HIF) pathway-associated mechanism, but the mode of action of LIP… Show more

“…Other pathways that play a role in the mechanosignaling processes were enriched after the US stimulation, such as those related to cell migration and regulation of PAK2p34, NOTCH4, and TNFR2/NFkB (Figure d). Interestingly, ITGA5, a protein involved in the regulation of focal adhesion and ECM organization was considerably up-regulated in the Nanocomp+US samples; ITGA activates FAK and thus the downstream signaling pathways PI3K/Akt, WNT, and MAPK, balancing cellular homeostasis between cell proliferation and cell survival …”

“…So, we evaluated the biological and chondrogenesis effects of CC-EVs in ADSCs. In addition, LIUS has been shown to alleviate chondrocyte damage in osteoarthritis [38] and enhance the regenerative potential of BMSC-derived extracellular vesicles in promoting cartilage regeneration [39]. Next, we aim to demonstrate whether our ultrasound-driven healing as an innovative method to enhance the production of chondrocyte-derived vesicles can enhance the ability of EVs to promote chondrogenesis in ADSCs.…”

“…In a simulative oxygen-deficient environment, through applying tandem mass tag (TMT) technology, it was observed that LIPUS increased the expression of chondrogenic factors, including sox9, collagen II and aggrecan, while reducing the expression of vascular endothelial growth factor (VEGF), which suggested that LIPUS can promote chondrocytes growth and inhibit angiogenesis in hypoxia-Induced TMJ-OA. Besides, the study also found that extracellular Matrix (ECM) proteins may become fresh treatment targets [97]. However, hypoxia contributes to activating the hypoxia-inducible factor pathway, and the expression of hypoxia inducible factor (HIF) existing in chondrocytes increases.…”
