The ATF6 branch of unfolded protein response and apoptosis are activated to promote African swine fever virus infection

Abstract: African swine fever virus (ASFV) infection induces apoptosis in the infected cell; however, the consequences of this activation on virus replication have not been defined. In order to identify the role of apoptosis in ASFV infection, we analyzed caspase induction during the infection and the impact of caspase inhibition on viral production. Caspases 3, 9 and 12 were activated from 16 h post-infection, but not caspase 8. Indeed, caspase 3 activation during the early stages of the infection appeared to be crucia… Show more

“…Reverse transcriptions PCR reactions (RT-PCR) were carried out using the SuperScript One Step RT-PCR (Life Technologies). Unspliced or spliced Xbp-1 mRNA was amplified as described [36]. Amplification of GAPDH mRNA was carried as a control for RNA extraction.…”

“…Activation of the ATF6 branch of UPR has been observed in cells infected by a handful of viruses including HSV-1 (Burnett et al, 2012), African swine fever virus (ASFV) (Galindo et al, 2012), West Nile virus (WNV) (Ambrose and Mackenzie, 2011) and HCV . Although ATF6 has been shown to promote the replication of WNV by suppressing interferon signaling (Ambrose and Mackenzie, 2013), the effect of ATF6 activation on replication of other viruses has not been studied in detail.…”

“…The primers used were: forward primer: 5 0 -CCTTGTAGTTGAGAACCAGG-3 0 and reverse primer 5 0 -GGGGCTTGGTATATATGTGG-3 0 . Amplification condition included an initial step of 45°C for 30 s, followed by a denaturation step of 94°C for 2 min, 30 cycles of 94°C for 30 s, 55°C for 30 s and 70°C for 30 s (Galindo et al, 2012). PCR products were resolved by electrophoresis in a 2% agarose gel and stained with GoldenView.…”

“…ASFV infection triggers ER stress and the unfolded protein response (UPR) in the host cells by inducing ATF6 signaling pathway of the UPR, but not PERK or IRE1 pathway. UPR regulation by ASFV might prevent early apoptosis to facilitate viral replication (Galindo et al, 2012). Tick-borne encephalitis virus infection activates the IRE1 and ATF6 signal pathways (Yu et al, 2013).…”
