The Association of MMP-9 Enzyme Activity, MMP-9 C1562T Polymorphism, and MMP-2 and -9 and TIMP-1, -2, -3, and -4 Gene Expression in Lung Cancer

Abstract: The frequencies of C1562T genotypes were found to be CC 67%, CT 30%, and TT 3% in the control group and CC 75%, CT 24%, and TT 1% in the patient group. It was determined that CC genotype frequency increases significantly in patients according to control group. Plasma MMP-9 enzyme activity levels increased in patients with lung cancer compared to the control group. The cut-off value of MMP-9 enzyme activity was determined as 7.76 ng/mL by receiver operating characteristics curve analysis. The sensitivity, speci… Show more

“…Genotypes for the C1562T polymorphism of MMP-9 were determined by polymerase chain reaction as described previously (Bayramoglu et al 2009) using the primers 5 0 -GCCTGGCACATAGTAGGC CC-3 0 (sense) and 5 0 -TTCCTAGCCAGCCGGCATC-3 0 (antisense). A 435-bp PCR product was cleaved with 1U SphI restriction enzyme (New England Biolabs, Ontario, Canada).…”

“…It degrades type IV collagen, proteoglycan, and laminin. MMP-9 is produced by tumoral cells as well as inflammatory cells such as neutrophil, eosinophil, monocyte, lymphocyte and alveolar macrophage (Chintala et al 1999;Corbel et al 2000;Apakkan Aksun et al 2001;Bayramoglu et al 2009Bayramoglu et al , 2011.…”

“…Matrix metalloproteinases (MMPs) have long been linked with cancer cell invasion and metastasis (15). Matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) are main members of MMPs and patients with high MMP-2 or MMP-9 expression have poorer overall survival (16)(17)(18). Nicotine increased the activity of MMP-2 in esophageal squamous…”

“…regions of other MMPs such as MMP-1 and MMP-9, may serve as promising markers for the prediction of lung cancer susceptibility and prognosis (30)(31)(32). From the molecular viewpoint, MMP-8 might increase cell adhesion by rearrangement of cytoskeleton actin, thus decreasing cell invasion (33).…”
