The association between N-methyl-d-aspartate receptor availability and glutamate levels: A multi-modal PET-MR brain imaging study in first-episode psychosis and healthy controls

Abstract: Background: Evidence from post-mortem studies and in vivo imaging studies suggests there may be reduced N-methyl-d-aspartate receptor (NMDAR) levels in the hippocampus in patients with schizophrenia. Other studies have reported increased glutamate in striatum in schizophrenia patients. It has been hypothesised that NMDAR hypofunction leads to the disinhibition of glutamatergic signalling; however, this has not been tested in vivo. Methods: In this study, we investigated the relationship between hippocampal NMD… Show more

“…We acquired data in the right striatum only because MRS studies have failed to find distinct hemispheric differences in glutamate ( Wood et al, 2008 ; Bustillo et al, 2011 ) and we had limited scanning time. Moreover, several studies have reported MRS glutamate findings in schizophrenia for the right striatum, so our choice of the right striatum enables comparisons with the existing evidence base ( de la Fuente-Sandoval et al, 2011 , 2013b ; Egerton et al, 2020 , 2023 ; Beck et al, 2022 ). Voxel placement and example spectra can be seen in Figure 1 .…”

“…The size of the detector in the longitudinal axis has been growing as a recent direction in improving PET physical functioning to boost scanner responsiveness [ 237 , 240 ]. Although this is by no means a simple procedure, it is a rather evident development; yet end users may experience practical difficulties when installing these bigger scanners [ 235 , 241 ].…”
