Cystic echinococcosis (CE) is one of the most widespread and harmful zoonotic parasitic diseases and it most commonly affects the liver. In this study, we characterized multiple changes in mouse hepatocytes following treatment with excretory-secretory (ES) products of Echinococcus granulosus protoscoleces by a factorial experiment. The cell counting kit-8 assay (CCK-8), the 5-ethynyl-2'-deoxyuridine (EdU) assay, and flow cytometry were used to detect the growth of hepatocytes. Inverted microscopy, scanning electron microscopy (SEM), and transmission electron microscopy (TEM) were used to observe the morphology and ultrastructure of hepatocytes. An automatic biochemical analyzer and an ELISA detection kit were used to determine six conventional hepatocyte enzymatic indices, the levels of five hepatocyte-synthesized substances, and the contents of glucose and lactate. Western blot analysis was conducted to analyze the protein expression of six rate-limiting enzymes in the glucose metabolism pathway in hepatocytes: glutamic-pyruvic transaminase (ALT), glutamic-oxalacetic transaminase (AST), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), gamma-glutamyl transpeptidase (GGT), and leucine arylamidase (LAP). The results of the CCK-8 and EdU assays both showed that ES could inhibit the proliferation of hepatocytes, and flow cytometry indicated that ES could promote apoptosis of hepatocytes. After ES treatment, the ultrastructure of hepatocytes was disrupted to a certain extent. The changes in the cell membrane and microvilli were observed through SEM, and the changes in the nucleus, mitochondria, and rough endoplasmic reticulum were observed through TEM. After ES treatment, the enzymatic activities of the six hepatocyte enzymes were increased in addition to the Fe metabolism and the synthesis of albumin (ALB), uric acid (UA), and urea, whereas the synthesis of transferrin (TRF) was decreased. The expression levels of all six key enzymes in the glucose metabolism pathway in hepatocytes were decreased, and the biological effects were significantly inhibited. We analyzed the causes and possible complications caused by various changes and advocate corresponding measures. We also propose possible mechanisms by which protoscoleces cause hepatocyte necrosis, but the specific mechanism requires further study.

Environmental condition is very important for the Dengue Fever Prevention. People in this area lacked the knowledge, awareness of an environmental condition. To achieve sustainable control of the occurrence of dengue fever, therefore, this mixed methodology research was performed: 1) to gather general information related to the community environment for dengue fever prevention, 2) to develop a paradigm for dengue fever prevention, and 3) to compare the levels of knowledge, attitude, and community participation after attending a training course on dengue fever prevention. The gathering general information related to the community environment for dengue fever prevention was performed by survey method, 341 participants were recruited by using the Taro Yamane formula and stratified random sampling, subsequently, the model was developed for training. After that, the training and evaluation were implemented. Thirty-two volunteers participated in the training on dengue fever prevention. The testing instruments consisted of Knowledge Test Form, Attitude Test Form, and Dengue Prevention Participatory Test Form. The results indicated that the dengue fever prevention paradigm was supported by the following six factors: 1) having reliable data, 2) providing training on disease prevention, 3) having competent agents, 4) presenting the dengue prevention system to the community, 5) having people participation, and 6) having environmental management. Concerning the training on dengue fever prevention, the villagersâ€™ post-test scores on knowledge, attitude, and participation in dengue fever prevention were significantly higher than their pre-test scores. This research increased the knowledge, attitude and participation of people after training on dengue fever prevention in their community.

The incidence of dengue in Latin America has increased dramatically during the last decade. Understanding the pathogenic mechanisms in dengue is crucial for the identification of biomarkers for the triage of patients. We aimed to characterize the profile of cytokines (IFN-γ, TNF-α, IL-1β, IL-6, IL-18 and IL-10), chemokines (CXCL8/IL-8, CCL2/MCP-1 and CXCL10/IP-10) and coagulation mediators (Fibrinogen, D-dimer, Tissue factor-TF, Tissue factor pathway inhibitor-TFPI and Thrombomodulin) during the dengue-4 epidemic in Brazil. Laboratory-confirmed dengue cases had higher levels of TNF-α (p < 0.001), IL-6 (p = 0.005), IL-10 (p < 0.001), IL-18 (p = 0.001), CXCL8/IL-8 (p < 0.001), CCL2/MCP-1 (p < 0.001), CXCL10/IP-10 (p = 0.001), fibrinogen (p = 0.037), D-dimer (p = 0.01) and TFPI (p = 0.042) and lower levels of TF (p = 0.042) compared to healthy controls. A principal component analysis (PCA) distinguished between two profiles of mediators of inflammation and coagulation: protective (TNF-α, IL-1β and CXCL8/IL-8) and pathological (IL-6, TF and TFPI). Lastly, multivariate logistic regression analysis identified high aspartate aminotransferase-to-platelet ratio index (APRI) as independent risk factors associated with severity (adjusted OR: 1.33; 95% CI 1.03–1.71; p = 0.027), the area under the receiver operating characteristics curve (AUC) was 0.775 (95% CI 0.681–0.869) and an optimal cutoff value was 1.4 (sensitivity: 76%; specificity: 79%), so it could be a useful marker for the triage of patients attending primary care centers.
