The Aryl Hydrocarbon Receptor Relays Metabolic Signals to Promote Cellular Regeneration

Abstract: While sensing the cell environment, the aryl hydrocarbon receptor (AHR) interacts with different pathways involved in cellular homeostasis. This review summarizes evidence suggesting that cellular regeneration in the context of aging and diseases can be modulated by AHR signaling on stem cells. New insights connect orphaned observations into AHR interactions with critical signaling pathways such as WNT to propose a role of this ligand-activated transcription factor in the modulation of cellular regeneration by… Show more

“…Restoration of butyrate, which is diminished in intestinal epithelial cells (IECs) after allo-HCT, improved IEC junctional integrity, decreased apoptosis, and mitigated GVHD ( 66 ). Aryl hydrocarbon receptor (AhR) is a cellular metabolic sensor ( 72 ). AhR ligands are derived from intestinal microbiota metabolism.…”

“…Their involvement in various cancer-associated processes is also well-known [ 66 , 67 ]. AHR is a ligand-activated transcription factor, which may interact with different pathways regulating cellular homeostasis including cellular regeneration in the context of aging and diseases [ 68 ]. In addition, AHR regulates CYP1B1 [ 69 ], which belongs to the cytochrome P450 superfamily of enzymes.…”

“…The characterization of this pathway indeed becomes more complex given the evidence that the role of AhR pathway in self-renewal is cell context and differentiation stage specific. 73,74 A phase I/II clinical study performed with one unmanipulated UCB unit and the progeny of the CD34 + cells from another UCB unit treated for 15 days ex vivo with SR1 demonstrated rapid neutrophil and platelet recovery. Importantly, this recovery was significantly more rapid than in patients receiving only the unmanipulated UCB.…”

“…Although this evidence clearly implicates the AhR pathway in HSC metabolism and hematopoiesis, additional studies are needed to precisely understand the role of the AhR in the self‐renewal of human HSCs. The characterization of this pathway indeed becomes more complex given the evidence that the role of AhR pathway in self‐renewal is cell context and differentiation stage specific …”
