Legume plants are able to establish a symbiotic relationship with soil bacteria from the genus Rhizobium, leading to the formation of nitrogen-fixing root nodules. Successful nodulation requires both the formation of infection threads (ITs) in the root epidermis and the activation of cell division in the cortex to form the nodule primordium. This study describes the characterization of RabA2, a common bean (Phaseolus vulgaris) cDNA previously isolated as differentially expressed in root hairs infected with Rhizobium etli, which encodes a protein highly similar to small GTPases of the RabA2 subfamily. This gene is expressed in roots, particularly in root hairs, where the protein was found to be associated with vesicles that move along the cell. The role of this gene during nodulation has been studied in common bean transgenic roots using a reverse genetic approach. Examination of root morphology in RabA2 RNA interference (RNAi) plants revealed that the number and length of the root hairs were severely reduced in these plants. Upon inoculation with R. etli, nodulation was completely impaired and no induction of early nodulation genes (ENODs), such as ERN1, ENOD40, and Hap5, was detected in silenced hairy roots. Moreover, RabA2 RNAi plants failed to induce root hair deformation and to initiate ITs, indicating that morphological changes that precede bacterial infection are compromised in these plants. We propose that RabA2 acts in polar growth of root hairs and is required for reorientation of the root hair growth axis during bacterial infection.

In plants, cell growth is dependent on vacuole enlargement, cell wall deposition and plasma membrane extension. Secretion (exocytosis) and uptake (endocytosis) of materials are two vital processes for cell function and cell–cell communication. The fusion of secretory vesicles with the plasma membrane (PM) during exocytosis is essential for proper cell function because it delivers proteins and lipids to the PM as well as proteins and polysaccharides to the cell wall. Basically, secretory proteins can be transported to the PM through the Golgi apparatus (conventional protein secretion) or bypassing the Golgi (unconventional protein secretion) via vesicle‐mediated pathways. In mature, nongrowing cells, the anterograde flow of materials is balanced by endocytosis, which retrieves excess membrane and recycles PM receptors and transporters. Clathrin‐mediated endocytosis (CME) and membrane microdomain‐associated endocytosis (MME) are the major well‐characterised routes. The balance of exocytosis and endocytosis is tightly regulated, which is crucial to both plant development and signalling processes. Key Concepts Conventional protein secretion (CPS) is the trafficking route for secretory proteins that are transported through the endoplasmic reticulum (ER), Golgi apparatus and (plasma membrane) PM via secretory vesicles. Unconventional protein secretion (UPS) is the process by which proteins are secreted by‐passing the Golgi apparatus. Endocytosis in plants shares conserved features with those in animals but also possesses some evolutionarily unique components. Clathrin‐mediated endocytosis (CME) and membrane microdomain‐associated endocytosis (MME) have been recognised to mediate the internalisation of cargo proteins. The balance of exocytosis and endocytosis regulates proteins and lipids in the PM and mediates signalling transduction and plant adaptation to an ever‐changing environment.

Differential regulation of aquaporins, small GTPases and V‐ATPases proteins in rice leaves subjected to drought stress and recovery

Mechanisms of drought tolerance are complex, interacting, and polygenic. This paper describes patterns of gene expression at precise physiological stages of drought in 35-day-old seedlings of Oryza sativa cv. Nipponbare. Drought was imposed gradually for up to 15 days, causing abscisic acid levels to rise and growth to cease, and plants were then re-watered. Proteins were identified from leaf samples after moderate drought, extreme drought, and 3 and 6 days of re-watering. Label-free quantitative shotgun proteomics resulted in identification of 1548 non-redundant proteins. More proteins were down-regulated in early stages of drought but more were up-regulated as severe drought developed. After re-watering, there was notable down regulation, suggesting that stress-related proteins were being degraded. Proteins involved in signalling and transport became dominant as severe drought took hold but decreased again on re-watering. Most of the nine aquaporins identified were responsive to drought, with six decreasing rapidly in abundance as plants were re-watered. Nine G-proteins appeared in large amounts during severe drought and dramatically degraded once plants were re-watered. We speculate that water transport and drought signalling are critical elements of the overall response to drought in rice and might be the key to biotechnological approaches to drought tolerance.
