The Arabidopsis male-sterile mutant dde2-2 is defective in the ALLENE OXIDE SYNTHASE gene encoding one of the key enzymes of the jasmonic acid biosynthesis pathway

Abstract: The Arabidopsis thaliana (L.) Heynh. mutant delayed-dehiscence2-2 (dde2-2) was identified in an En1/Spm1 transposon-induced mutant population screened for plants showing defects in fertility. The dde2-2 mutant allele is defective in the anther dehiscence process and filament elongation and thus exhibits a male-sterile phenotype. The dde2-2 phenotype can be rescued by application of methyl jasmonate, indicating that the mutant is affected in jasmonic acid biosynthesis. The combination of genetic mapping and a c… Show more

“…In addition the sequence of the ps-2 gene will give us more insights into the physiology of anther dehiscence in tomato. Similar phenotypes have been observed in some Arabidopsis mutants where, in most of the cases, the jasmonic acid pathway was involved (Feys et al 1994;Ishiguro et al 2001;von Malek et al 2002;McConn and Browse 1996;Park et al 2002;Sanders et al 2000;Stintzi and Browse 2000;Xie et al 1998). Once the ps-2 gene is cloned, it will be interesting to know whether this gene is ortholog to one of those genes already identiWed in Arabidopsis.…”

“…The molecular mechanism of ps-2 in tomato is unknown, but similar mutant phenotypes have been observed and studied mainly in Arabidopsis, such as the mutant myb26 (Steiner-Lange et al 2003), the mutants coi1 (Feys et al 1994;Xie et al 1998) and dad1 (Ishiguro et al 2001), the double mutants opr3/ dde1 (Sanders et al 2000;Stintzi and Browse 2000) and aos/dde2-2 (von Malek et al 2002;Park et al 2002) and the triple mutant fad3/fad7/fad8 (McConn and Browse 1996). They are characterized by defects in Wlament elongation, timing of anther dehiscence and often show reduced pollen viability.…”

“…In our analysis, we use the JA-biosynthesis mutant aos, in which the single Allene Oxide Synthase (AOS) gene in Arabidopsis has been knocked out (Park et al, 2002) and the production of JA and its precursors is impaired (von Malek et al, 2002). In the aos mutant, JA downstream signaling can be rescued by exogenous JAs.…”

“…Unlike aim1 (Richmond and Bleecker, 1999), floral morphology was normal in cts mutants, although they exhibited delayed filament extension (Fig. 1B), a phenotype that is shared by plants lacking functional allene oxide synthase and COI1, which are required for JA biosynthesis and perception, respectively (Feys et al, 1994;Park et al, 2002;von Malek et al, 2002). However, application of JA did not rescue the anther extension phenotype of cts flowers (data not shown).…”
