The Aquaporin 3 Promoter Polymorphism −1431 A/G is Associated with Acute Graft Rejection and Cytomegalovirus Infection in Kidney Recipients Due to Altered Immune Cell Migration

Abstract: Major complications after kidney transplantation are graft rejection and cytomegalovirus (CMV) infection, which are related to T-cell function, which depends on aquaporin 3 (AQP3) expression. The impact of the AQP3 A(−1431)G promoter polymorphism in kidney transplant recipients was unelucidated and we explored the effect of AQP3 polymorphism on immune cell function and its association with graft rejection and CMV infection in 237 adult patients within 12 months after transplantation. AQP3 promoter polymorphism… Show more

“…However, we observed a significantly higher incidence of CMV infection among middle-aged patients (31–50 years). Our results concerning gender agreed with previous studies; however, several studies have shown no association between age and the risk of CMV infection among the post-renal transplant recipients [ 1 , 7 , 8 ]. Our finding could be attributed to the higher proportion of middle-aged individuals in our sample pool.…”

“…Several studies have reported no significant correlation between preoperative diabetes and risk of CMV infection [ 7 , 8 ]. Another study conducted by Beyler et al also showed a limited association between preoperative diabetes and postoperative CMV incidence; however, they attributed their results to a small number of patients and a short follow-up period [ 14 ].…”

“…Here, AQP3 regulates T-cell migration by mediating the transport of hydrogen peroxide [18]. As we have also demonstrated in the past, a polymorphism in the AQP3 gene is associated with T-cell migration [19], and altered T-cell migration could play a role in sepsis patients. Furthermore, AQP3 mediates both chemokine production and T cell trafficking [20].…”

“…AQP3 is involved in T-cell function, and a different AQP3 expression alters T-cell migration [19,30]. Hence, in our study, an altered T-cell function could impact sepsis survival.…”

“…Clinical manifestations can range from mild to severe life-threatening diseases [ 4 ] and can present as a non-specific systemic viral disease [ 5 ] or a localized tissue-invasive infection, most commonly including pneumonia [ 6 ], hepatitis [ 7 ], colitis [ 8 ], and retinitis [ 9 , 10 ]. The diagnostic of human cytomegalovirus plays an important role in the follow-up and treatment of transplant patients because it is a cause of graft rejection [ 11 ]. Different studies report the diagnostic methods of human cytomegalovirus, such as the serological methods that determine the presence of IgG antibodies, which is an indicator of infection in the past and of IgM for recent infection [ 12 ].…”
