The application of multiple linear regression to the measurement of the median particle size of drugs and pharmaceutical excipients by near-infrared spectroscopy

Abstract: A number of powdered drugs and pharmaceutical excipients were used to demonstrate the ability of near-infrared spectroscopy to measure median particle size (d50). Sieved fractions and bulk samples of aspirin, anhydrous caffeine, paracetamol, lactose monohydrate and microcrystalline cellulose were particle sized by forward angle laser light scattering (FALLS) and scanned by fibre-optic probe FT-NIR spectroscopy. Two-wavenumber multiple linear regression (MLR) calibrations were produced using: NIR reflectance; a… Show more

“…NIRS has also been successfully used in a variety of other analytical chemistry applications such as the detection of degradation products in tablets (7); studying drug moisture content over time and water mobility within the drug crystal lattice (8,9); real-time monitoring of moisture during processing using fluidized bed granulation (10,11) or roller compaction (12), measurement of particle size, and size distribution (13)(14)(15); and determining the tablet drug content and content uniformity (16)(17)(18)(19)(20)(21)(22)(23).…”

“…Vice versa, absorbance bands constitute a complicating factor for the determination of physical properties of the solid. Several studies have investigated the particle size in various processes (6,8,28,(30)(31)(32)(33) and the moisture content of lyophilized sugars and proteins by NIR (7,8,30,(34)(35)(36). However, NIR has seldom been applied to monitor the spray-drying process, and has rarely been used when proteins are the model system (37).…”
